Zhizi(Fructus Gardeniae) originates from the fruits of Gardenia jasminoides Ellis, and is recorded by China Pharmacopeia. Its main components are iridoid glycosides and crocins. In China Pharmacopeia (2000, part 1), geniposide is the criteria component for quantifying, but there has no quality standard of crocins, an objective evaluation of Fructus Gardeniae and an effective control of its preparations can't be done. To consummate the quality standard of Fructus Gardeniae and to provide a scientific quantitative method of Gardeniae Yellow, Fructus Gardeniae was studied on its quality control method and chemical constituents. The studies included as here:1. Four pharmacognosy verifications were studied of Zhizi, Sh-anzhizi and Shuizhizi who are used as "Zhizi" in Chinese Medicinein different areas. The results indicated that Zhizi, Shanzhizi and Shuizhizi had no differences except some differences in size. The results accord with what Zhong Guo Zhi Wu Zhi reports.2. For the first time, the method of switching UV detective wavelength was established to detect iridoid glycosides and crocins of different detective wavelength in Fructus Gardeniae simultaneously. This method is simple with good repetition, and can be used for the quality control method of Fructus Gardeniae.2. For the first time, the finger print spectrum of Gardenia jasminoides Ellis from GAP base of operations in Naxi district of Luzhoucity, Sichuan province was established. 10 batches of Fructus Gardeniae from the GAP base of operations were analyzed. With a reference peak of geniposide, 19 peaks have been identified as common ones. In addition, the finger print spectrum was applied to analyze 10 batches of Fructus Gardeniae from different areas. The results were that all the samples showed high similarities(>0.900). The result suggests that this finger print spectrum can well reflect the characteristics of Fructus Gardeniae.4. Using HPLC method, 10 batches of Fructus Gardeniae from different producing areas were analyzed, and the contents of geniposide and crocins were obtained. The results showed that the content of geniposide had no obvious relationship with crocins, and that the content of crocin-1 had a direct ratio to the content of crocins.5. The contents of geniposide and crocins of Fructus Gardeniae of different develop periods were for the first time determined simultaneously. The result showed that the highest content periods of geniposide and crocins were not coincident. Determining the best collection periods should base on the chemical component after the maturity of Zhizi.6. Seven compounds were isolated from the 70% ethanol extract of Fructus Gardeniae. Their structures were elucidated as picrocrocinic acid(b), geniposide(c), crocin-l(d), 13-cis-crocin-l(e), crocin-2(f) and crocin-3(g) by spectrum techniques, such as TLC\ ESI-MS> 'H-NMR>13C-NMR, and 13-cis-crocin-l is firstly isolated from the plant.
