Objective:(1) To establish a simple, feasible and stable animal model of pulmonarymetastasis of nasopharyngeal carcinoma cells;(2)to compare pulmonary metastases with subcutaneous transplantation tumor KiSS-1mRNA expression differencesMethods:(1) By using the concentration of2x10~6/L,5x10~6/L, and10x10~6/LSUNE-1nasopharyngeal carcinoma cells axillary subcutaneous inoculation in nudemice, mice were euthanized at timely anatomy and record the metastatic tumor weight,the number and size of lung metastases.(2) By using the RT-PCR compare pulmonary metastases with subcutaneoustransplantation tumor KiSS-1mRNA expression differences.Results:(1) at a concentration of2×10~6/L,5×10~6/L,10×10~6/L of0.2mlSUNE-1nasopharyngeal carcinoma cells in nude mice inoculated subcutaneouslyaxilla, three groups could be a tumor, tumor time was10.50±3.02days,11.30±2.21days,10.20±2.69days, no statistically significant difference (P>0.05), three groupsof tumor weight was1.269±0.482g,1.683±0.436g,2.183±0.226g, there aresignificant differences among the three sets of data (P <0.05), three groups of lungmetastases formation rate was70%,60%,70%, respectively, no significant difference(P>0.05), the number of lung metastases were26.40±5.57,44.02±7.12,44.77±7.00,statistics show that2×10~6/L concentration group with5×10~6/L group,10×10~6/L group were significantly different (P <0.05), and5×10~6/L group and10×10~6Lwas no statistical difference.(2) RT-PCR comparison of pulmonary metastases and subcutaneous tumorKiSS-1mRNA. Two organizations were amplified KiSS-1mRNA, lung metastasesexpressing KiSS-1gene mRNA levels below xenograft expression (P <0.05). Conclusion:The experiment to establish a stable lung metastasis of nasopharyngeal carcinoma innude mice model of nasopharyngeal carcinoma research and KiSS-1gene provides animportant animal model. Cell lung metastases KiSS-1gene expression at the mRNAlevel below the subcutaneous tumor expression, indicating that KiSS-1gene plays animportant role in the metastasis of tumors.
