| Objective:(1) To clarify the role of Ki SS-1 in nasopharyngeal carcinoma invasion and metastasis.(2) To evaluate the application value of EPCs transfected Ki SS-1 gene in theprevention and therapy of nasopharyngeal carcinoma.Methods:(1) To construct the recombinant plasmid Ki SS-1 and transfected EPCs isolated and cultured from human umbilical vein, access to high expression of Ki SS-1gene stably transfected cells(Ki SS-1-EPCs);(2) Construction of a nude mouse model of NPC and injected Ki SS-1-EPCs through its tail vein;(3) To observe the influence of Ki SS-1-EPCs in tumor and its metastases.Results:The human umbilical cord blood progenitor cells were obtained successfully by density gradient centrifugation and 6% hydroxyethyl starch sedimentation, and completing their training, passage, amplification and identification; The Ki SS-1 gene was transfected into human umbilical cord endothelial progenitor cells completely,and endothelial progenitor cells transfected were identified by western blot method; A stable model of lung metastasis of nasopharyngeal carcinoma was established in nude mice;The EPCs of transfected Ki SS-1 plasmid(Ki SS-1-EPCs) was injected into nude mice with nasopharyngeal carcinoma via the tail vein of mice. Each rat was injected the cell for the concentration of 2×106/m L,and as experiments group: then EPCs of no transfected Ki SS-1 recombinant plasmid in the same concentration was injected into the tail vein of nude mice with nasopharyngeal carcinoma, and take it as a control group. The results showed that the ransplanted tumor weight of experimental group and the control group were 1.385±0.207 g, 1.474±0.229 g, no significant difference(P> 0.05) between the two sets of data.The two groups of lung metastases formation rate is 5%, respectively, 30%, a significant difference(P<0.05), the numberof lung metastases were 19.00 and 30.02±9.09.Conclusion:In this study,a stable model of lung metastases of nasopharyngeal carcinoma in nude mice was established, and it provides an important animal model for nasopharyngeal carcinoma and Ki SS-1 gene research. EPCs transfected Ki SS-1 gene can inhibite the lung metastasis of nasopharyngeal carcinoma in nude mice. |
