Objective: To investigate the effects of different fluid restriction resuscitation on Tlymphocytes and IL-6,TNF-α in rats with hemorrhagic shock. Methods: All the healthy SD rats were randomly divided into four groups: normalcontrol group, sodium lactate Ringer’s fluid resuscitation group(crystal group),hydroxyethyl starch resuscitation group(colloidal group), crystal glue fluid resuscitationgroup(crystal gel group). The model of hemorrhagic shock were established in theexperiment groups. Draw blood of rats in each experiment group at the time of beforeshock,MAP=40mmHg maintain30min (0time point),MAP=60mmHg maintain30min(1time point),MAP=80mmHg maintain60min(2time point),and24hours laterafter successful resuscitation(3time point). The normal control group rats were drewblood only before the experiment and the3time point. Determination of CD4+Tlymphocyte, CD8+T lymphocyte number and the ratio of CD4+/CD8+using flowcytometry technique.ELISA method was used to determine IL-6and TNF-α content inplasma.Results:1.Before shock and the shock began to recove(MAP=40mmHg maintain30min,namely0time point), the CD4+T lymphocyte number, CD8+T lymphocyte number and the ratioof CD4+/CD8+and the contents of IL-6and TNF-αin plasma of each group rats showedno statistical significance(P>0.05).2.In the process of limited resuscitation, the number of CD4+T lymphocyte and the ratioof CD4+/CD8+in crystal group were decreased and CD8+T lymphocyte number wercreased and then decreased. The content of CD4+T lymphocyte and the ratio ofCD4+/CD8+in crystal group were higher than that of the control group at the3timepoint,the difference showed statistical significance(P<0.05). The number of CD4+Tlymphocyte and the ratio of CD4+/CD8+in crystal gel group were higher than that of the colloidal group and at the1time point,2time point and3time point, the difference wasstatistical significance(P<0.05).3.In the process of limited resuscitation, the content of IL-6and TNF-αin crystal groupwere increased firstly and then decreased. The content of IL-6and TNF-α incrystal group were higher than that of the control group at the3time point, thedifference showed statistical significance(P<0.05). The content of IL-6and TNF-αincolloidal group and crystal gel group showed a declining trend. The content of IL-6and TNF-αin colloidal group and crystal gel group were lowed than that of the controlgroup at the3time point, the difference was statistical significance(P<0.05). The contentof IL-6and TNF-αin crystal gel group were lowed than that of the colloidal group andat the1time point,2time point and3time point, the difference was statisticalsignificance(P<0.05).Conclusion:1. Shock induced immune disorders. compared with crystalloid and colloid, inhibitionof immune disorders using crystal glue can be better.2.In the process of limited resuscitation, crystal glue liquid can better inhibit the releaseof proinflammatory mediators. Crystal glue liquid may bea relatively better resuscitate liquid.
