Koraiensis corns,as byproducts of agricultural products processing industry, were once ignored by people. In recent years,people became aware of that the resource which was been wasted, had important physiological activity with further research.The effects on immunomodulatory and cancer treatment of Koraiensis corns are huge, and polysaccharide is the main active ingredient of them.The analysis, preparation and biological activity of Pinus koraiensis corn polysaccharide was studied in this research.The main innovation as follows:1Aconent analysis method-3,5-dintro salicylic acid colorimetic method(DNS method) was established to determine the conent of Pinus koraiensis corn polysaccharide,and this was attempted firsrly in this experiment. Compared with other methods, the DNS method not only didn’t use sulfuric acid, and the reagents used were stabile,but also thtat it could be measured directly after removing the interference of monosaccharide composition,so the result was more accurate. While trying to create a DNS method for the determination of Pinus koraiensis corn polysaccharide, reaction conditions and method investigation were studied and the results showed that the method was simple, accurale,reproducible and can be used for the determination of Pinus koraiensis corns polysaccharide.2The membrane separation technology was firstly used to preparating Pinus koraiensis corn polysaccharide,and the process was founded.Firstly,the boiling method was used to extract the polysacchairide.After concentration step-by-step, activated carbon absorption and other steps were used to remove the impurity of the feed liquid.Then micro filtration and ultrafiltration was used to separating and purificating. During the experiment, the best conditions of pretreatment was found. And effect of concentration,temperature,pressure of feed on exudate flux of MF was studied,and the best condition was found by orthogonal test. The best condition of MF was25g/L,0.10MPa,40℃.Finally, ultrafiltration was used to separating and purificating.After UF,the concentration of polysaccharide was44.70%,5times higher than7.2%of before.3Membranes,with different molecular cut off were used to separated MF permeance of Pinus koraiensis corn polysaccharide. Polysaccharide with different molecular weight was preparated.The content of polysaccharide was determined respectively, and the weight distributing of Pinus koraiensis corn polysaccharide was studided. Polysaccharide with molecular weight>100kDa accounting for9.50%,50-100kDa accounting for70.20%,10-50kDa accounting for3.20%,6-10kDa accounting for17.1%.The first hand information was gotten.4The antibacterial activity of Pinus koraiensis corn polysaccharide was studied firstly. The results showed that,the polysaccharide could inhibit the activity of E. coli, Penicillium and Saccharomyces cerevisiae,and the inhibition of Penicillium was most strong. There was no significant inhibition of Staphylococcus aureus. The polysaccharide solution on the minimum inhibitory concentration (MIC) of E. coli are0.5mg/mL; minimum inhibitory concentration (MIC) of Penicillium and Saccharomyces cerevisiae0.25mg/mL. Pinus koraiensis corn polysaccharide without protein had weaker bacteriostatic activity,compared with polysaccharide with protein. It was also found that Pinus koraiensis corn polysaccharide with different molecular weight had different bacteriostatic activity.Order as follow:>100kDa、50kDa-100kDa、10kDa-50kDa, and the Pinus koraiensis corn polysaccharide with molecular weight6kDa-10kDa had little bacteriostatic activity. Pinus koraiensis corn polysaccharide, molecular weight>100kDa and between50kDa-100kDa,had most strong bacteriostatic activity to Penicillium,than Saccharomyces cerevisiae and E. coli.That was, its bacteriostatic activity to fungi Russian more strong than to bacteria.Pinus koraiensis corn polysaccharide molecular weight10kDa-50kDa, had most strong bacteriostatic activity to E. coli than Penicillium and Saccharomyces cerevisiae. That was, its bacteriostatic activity to bacteria was more strong than to fungi Russian.Above all the innovation, in this experiment, the process of ultrafiltration and alcohol precipitation to preparating Pinus koraiensis corn polysaccharide was compared. Results showed that the content of Pinus koraiensis corn polysaccharide preparated by ultrafiltration was44,70%, much higher than23.88%of alcohol precipitation product. In addition, the comparison of ultrafiltration and alcohol precipitation process in the cost of the process route, showed that the ultrafiltration had more advantages. Ultrasonic extraction method was also studied in this research,and result showed that compared with hot water boiling method, Ultrasonic extraction method could give higher yield and content of polysaccharide.Analysis, preparation, purification and biological activity of the Pinus koraiensis corn polysaccharide was researched exploringly, and remarkable progress had been made. I hope that the research can provide reference for the the related in-depth study.
