PART ONEEFFECT OF AMBROXOL ON THE POLYSACCHARIDEINTERCELLULAR ADHESIN OF STAPHYLOCOCCUSEPIDERMIDIS BIOFILM IN VITROObjectiveTo explore the effect of ambroxol on polysaccharide intercellularadhesion(PIA) of Staphylococcus epidermidis biofilm(BF) modelestablished in vitro.MethodsThe mature BF of Staphylococcus epidermidis was established afterincubated for24h on the slide. With different concentration of ambroxolintervention for24h, the BF was oscillated down and collected forinvestigation. The effect of ambroxol on PIA was detected through the sulfuric acid-phenol method.RusultsPIA content(μg/10mg) of the1.875mg/ml ambroxol treatment groupand control group were (345.1050±8.9603) and (403.6269±16.9943),respectively, t=7.213,P<0.05.The effect of ambroxol with theconcentration of3.75mg/ml had the same impact but more obvious.ConclusionAmbroxol can decrease the content of PIA, which effect waspositively relevant to the concentration of ambroxol. With different concentration of ambroxolintervention for8h, the BF was oscillated down and extracted for RNA.The effect of ambroxol on the mRNA expression of genes which areimportant to the synthesis of PIA such as icaA, icaR, δBand luxS weremeasured by reverse transcription PCR.RusultsThe mRNA expression of icaA (OD/control) of1.875mg/mlambroxol treatment group and control group were (0.4084±0.0461) and(0.5540±0.0341),respectively, t=6.008, P<0.05;The mRNAexpressionof icaR (OD/control)were (0.4253±0.0513)and(0.2826±0.0636),respectively, t=13.066,P<0.05;The mRNA expression of δB(OD/control)were (0.4236±0.0258) and (0.5291±0.0246), respectively, t=6.968,P<0.05;The mRNA expression of luxS (OD/control)were(0.2947±0.0367) and (0.1783±0.0385), respectively,t=5.180,P<0.05.The effect of ambroxol with the concentration of3.75mg/ml had the same impact but more obvious.ConclusionAmbroxol can decrease the mRNA expression of icaA and δBandincrease the mRNA expression of icaR and luxS, which effect waspositively relevant to the concentration of ambroxol.
