Objective:To construct Octamer-binding transcription factor-4adeno-virus eukaryotic expression vector pAV.Ex1d.–CMV>mOCT4/IRES/eGF-P, and study its effect on myocardial regeneration.Methods: Gateway Technology was used to construct eukaryotic expr-ession vector pAV.Ex1d.–CMV>mOCT4/IRES/eGFP and packaged thevector with adenoviruses. After adenovirus packaging,injected it into themouse myocardium by microliter syringe. The expression of OCT4wasdetermined by the methods of immunofluorescence and reverse transcri-ption-polymerase chain reaction (RT-PCR),hematoxylin-eosin(HE) staini-ng was used to detect whether the myocardial tissue was changed after theexpression of OCT4.Constructed mouse myocardial infarction model andused the hematoxylin-eosin(HE) staining to identify the infarct area,theninjected the OCT4Adenovirus Vector into the area surroundings in theinfarct area,and detected the expression of Cardiac Troponin T by westernblot after four weeks.Results:OCT4could be detected in the Myocardial cell nucleus and showed redfluorescence, but could not be detected in the control group andthe the control virus group. RT-PCR could detect the expression of OCT4in OCT4Adenovirus Vector group but notin the control virus group and thecontrol group.There was no pathological change in the myocardial tissueafter OCT4expressed in vivo.4weeks after injecting the adenovirus vectorinto the infarct area,the expression level of Cardiac Troponin T in OCT4group had obvious differences compared to the control group and thecontrol empty virus group(p<0.05).Conclusion: Exogenous OCT4can be expressed in the maturemyocardial tissue in vivo and promote myocardial regeneration...
