Objective:Hemangioma is a common congenital benign tumor in Neonate, mainly comes from the residual embryos in the process of embryonic development or vascular endothelial cells, Hemangioma is divided into true hemangioma and vascular malformation. Hemangioma has the characteristics of abnormal vascular endothelial cell proliferation, the oral and maxillofacial are attacked most probablely, the most hemangioma can regress along with the age, but nearly20%of hemangioma can’t fade. A simple solutions effective for hemangioma is still not found by now. In2008, the French doctor Leaute-Labreze discovered propranolol can promote hemangioma faded, but so far the mechanism is unclear.the experiment used human umbilical vein endothelial cells (HUVECs) as the model, test the effect of propranolol on vascular endothelial cell growth、 apoptosis、and growth factor expression in vitro. Caspase3is the core control protein of apoptosis in the Vascular endothelial cell, Vascular endothelial growth factor (VEGF) involved in promoting proliferation and migration of endothelial cells, increased Vascular permeability; VEGF has been identified as the main cause of angiogenesis, it stimulates endothelial cell proliferation, migration and tube formation; So this study attempts to study the effect of propranolol on Human umbilical vein endothelial cells (HUVEC) survival, changes of Caspase3and vascular endothelial growth factor expression, and use the classical antitumor drug pingyangmycin as a positive, in order to syudy the mechanism of propranolol treatment of hemangioma and provide theoretical basis for treatment prospects. Methods:HUVECs were obtained from the Typical species preservation Center of Wuhan University. HUVECs were incubate to the fourth generation or the fifth generation as the research specimens, culture umbilical vein endothelial cells with propranolol and PYM Respectively, compare the cell survival and proliferation ability under the action of the two drugs; test by MTT method to detect the cell survival rate, Western Blot detect the expression of apoptosis protein Casepase-3; ELISA detect the expression changes of VEGF; The statistical significance of raw data between the groups in each experiment was evaluated using unpaired Student’s t-test or ANOVA followed by Student-Newman-Keuls multiple comparison post-test With SPSS16.0. P value<0.05was considered as statistically significant. The results were expressed as means±SEM of the indicated n values.Results:1> there was no significant difference among Control group, PYM group and propranolol group in48h (P>0.05); In24h, there was significant difference among the groups (P<0.01), drug groups were significantly inhibited in cell proliferation, and PYM group was obviously higher than that of propranolol,in propranolol group the inhibition rate obviously increased with the increase of drug concentration,in PYM, there was no obvious correlation betweenthe inhibition rate and the concentration of PYM.2、The apoptosis protein Caspase3are all expressed in every group, the expression of the PYM group is the highest, the control group is the least.3, the VEGF expression have obvious difference among the groups (P<0.05), propranolol and PYM can significantly inhibit the expression of VEGF,and there is no difference between propranolol and PYM.Conclusion:In the action of propranolol, the cell proliferation ability and VEGF protein expression of HUVEC are all decreased obviously, the apoptosis protein Caspase-3’s secretion is increased; The results show that propranolol can inhibit the growth of endothelial cells, which has certain therapeutic effects on neonatal hemangioma, inhibition of vascular endothelial growth factor VEGF protein expression and promote apoptosis protein expression of Caspase3belong to some of its mechanism of action.
