Objective: To investigate the association of Fetuin-A gene rs1071592,rs2248690and rs2593813single nucleotide polymorphisms (SNPs) withpolycystic ovary syndrome (PCOS) in Chongqing Han population.Methods: A total of147PCOS patients and156healthy controls weregenotyped by TaqMan SNP Genotyping Assay.Hyperinsulinemic-euglycemic clamp was performed in147PCOS womenand20controls.Results: A significantly increased frequency of the homozygousrs1071592AA genotype (P_c<0.05) and A allele (P_c<0.05) were observedin PCOS patients as compared with controls. There was no significantdifference in genotype and allele frequencies of rs2248690, rs2593813between PCOS patients and normal controls (P_c>0.05). Binary logisticregression analysis showed that women with AA genotype of rs1071592were4.93times more likely to develop PCOS (OR4.933,95%CI1.593-15.278, P<0.05), compared with women with CC genotype ofrs1071592. In addition, dominant model analysis showed that AA+AC genotype carriers of rs1071592had higher levels of BMI, fasting seruminsulin, HOMA2-IR and lower M value compared with CC genotypecarriers of PCOS (P<0.05). Recessive model analysis showed that PCOSpatients carrying AA genotype of rs1071592had higher levels of fastingserum insulin, HOMA2-IR and lower M value compared with PCOS womencarrying AC+CC genotype (p<0.05).Conclusions: People with SNPs variants of rs1071592in Fetuin-Agene may have an increased susceptibility to PCOS and the A allele ofrs1071592might be a predictive marker of PCOS.
