Objective: To observe the transfection of monoacylglycerol lipaseshRNA (MAGL-shRNA) and the effect of inhibiting metastasis ofhepatocellular carcinoma in rat by using ultrasound-targeted microbubbledestruction (UTMD).Methods: Liver tumor models of rats were established. The successfulestablishment of tumor was testified by pathologic biopsy andtwo-dimensional ultrosound.40rats were randomly divided into4groups:phosphate-buffered salilne group, lipid microbubbles loaded MAGL-shRNA(MAGL-shRNA+MB) group, pure lipid microbubbles+ultrasound (MB+US)group, and lipid microbubbles loaded MAGL-shRNA+ultrasound(MAGL-shRNA+MB+US) group. The microbubbles containingMAGL-shRNA plasmid were injected through the tail vein of rat1ml.Ultrasound gene transfection instrument was applied on MB+US andMAGL-shRNA+MB+US groups for irradiation after the target gene inject,with the radiation frequency of1kHz, sound intensity of2W/cm2, and uesd pulse irradiation method20min, with the interval time of10s. Theexpression of MAGL protein was detected by Western-blot. Proteinexpression of MAGL, matrixmetalloproteinase-2(MMP-2), CD34, andproliferating cell nuclear antigen (PCNA) was detected byimmunohistochemistry (IHC). Survival time and tumor metastasis rate ofrats were compared.Results: The protein expression of MAGL in HCC tissue was higherthan in normal tissue (P<0.05). MAGL-shRNA could be injected preciselyinto tumor tissue and targeted release, the expression of MAGL protein wasfound in all HCC tissue. Expression in MAGL-shRNA+MB+US group waslower than others (P<0.01). The protein expression of MMP-2, CD34, andPCNA detected by IHC in MAGL-shRNA+MB+US group was lower thanothers (P<0.01). Survival time of rats in MAGL-shRNA+MB+US groupwas longest(P<0.05). Tumor metastasis was found in all groups, tumormetastasis rate was lowest in MAGL-shRNA+MB+US group (P<0.05).Conclusions: Ultrasound targeted microbubble destruction caneffectively transfect MAGL-shRNA into target tissues and play a inhibitioneffect of metastasis in liver cancer.
