Objective: To investigate the expression of WASP-familyverprolin-homologous protein1(WAVE1)、Matrix Metalloproteinase-2(MMP-2)andMatrixMetalloproteinase-9(MMP-9)inepithelial ovariancancer(EOC)、benign ovarian tissues and normal ovarian tissues, andcombined with patients’ clinical pathological data, to explore the clinicalsignificance of WAVE1、MMP-2and MMP-9in epithelial ovarian cancer.Methods: Fourty-seven epithelial ovarian cancer and thirty benignovarian tissues were collected from February2010to November2011inthe First Affiliated Hospital of Chongqing Medical University. and thecontrol group (normal ovarian tissues from the hysteromyoma or earlycervical cancer patients who underwent operation,n=21).1. The expressions of WAVE1, MMP-2and MMP-9in47cases ofepithelial ovarian cancers,30ovarian benign tumors and21normal ovariantumors were detected by SABC immunohistochemistry method.Thecorrelation analysis was done among WAVE1, MMP-2and MMP-9;2.WAVE1、MMP-2and MMP-9mRNA level was determined by RT-PCR.Results:1.Immunohistochemistry(IHC) identified that the positiveexpressions of WAVE1in epithelial ovarian cancer,benign ovarian tissuesand normal ovarian tissues were74.47%,16.67%,4.76,respectively;while MMP-2in three groups were82.98%,33.33%,14.29%;MMP-9were74.47%,20.00%,4.76%,The positive expression rates of WAVE1,MMP-2and MMP-9in epithelial ovarian cancer were significantly higherthan ovarian benign tumors and normal ovarian tissues(P<0.05).2. Spearman correlation analysis showed that positive correlation werefound between WAVE1and MMP-2,MMP-9in epithelial ovarian cancer(r1=0.773,P<0.05;r2=0.664,P<0.05).3. The expressions of WAVE1were correlated to clinicalstage,histological differentiation and Ca-125value(P<0.05), not correlatedto age and the size of tumor(P>0.05).While the expressions of MMP-2、MMP-9were related to clinical stage(P<0.05), not related to age,histological differentiation,Ca-125and the size of tumor(P>0.05).4. The data of RT-PCR revealed that the mRNA levels of WAVE1inepithelial ovarian cancer,benign ovarian tissues and normal ovariantissues were0.75±0.10、0.32±0.05、0.32±0.04,respectively, MMP-2mRNA levels in each group were0.73±0.04、0.41±0.11、0.37±0.09,MMP-9mRNA levels in each group were0.75±0.07、0.43±0.08、0.38±0.05,By statistical analysis, The mRNA levels of WAVE1, MMP-2and MMP-9in epithelial ovarian cancer weresignificantly higher than ovarian benign tumors and normal ovariantissues(P<0.05).Conclusion: WAVE1,MMP-2and MMP-9are highly expressed inepithelial ovarian cancer,which reveals that they may involve in thedevelopment of EOC.Packing screening of WAVE1,MMP-2and MMP-9in the early phase may have crucial clinical significance for evaluation ofprogress and prognosis of epithelial ovarian cancer.
