| Objective: To explore the role of CC-chemokine ligand18(CCL18) inthe proliferation,migration and chemoresistance of a nasopharyngealcarcinoma cell(HNE-1),and investigate the clinical significance ofCCL18in nasopharyngeal carcinoma.Method: Human nasopharyngeal carcinoma cell line HNE-1were usedfor this study. Recombinant chemokine CCL18were cultured with HNE-1together to test the cell proliferation by MTT assay and examine thechemotaxis of tumor cells through Transwell zeta. Different concentrationsof cisplatins were added in tumor cells cultured with recombinantchemokine CCL18to amount of the Median Lethal dose (LD50) of cells byMTT assay.Result: Recombinant CCL18had no effect on HNE-1cells growth.Transwell zeta method revealed that different concentrations of recombinantCCL18promoted the migration ability of NPC cells(all P<0.05),andobserved a clear-cut concentration-dependent effect of CCL18. CCL18modulated the sensitivity of HNE-1cells to cisplatin treatment, and reduced the influence of cisplatin.Conclusion: CCL18could be used as a biomarkers of NPC.CCL18play an important role in migration and chemoresistance of NPC,and havecertain clinical significance in diagnosis and treatment of NPC. |
