Objective:To explore the expression of TRIM29in the high metastatic potential nasopharyngeal carcinoma (NPC) cell line5-8F and lower metastatic potential NPC cell line6-10B and to investigate the effect of TRIM29expression on NPC cell proliferation, cycle distribution, migration and invision, so as to know the relationship between TRIM29and NPC, to study the role of TRIM29in the invasion and metastasis of NPC cells.Methods:(1) Quantitative Reverse Transcription Polymerase Chain Reaction (qRT-PCR) and Western Blotting was respectively used to detect the mRNA levels and the protein expressional levels of TRIM29in5-8F cells and6-10B cells;(2) Liposome transfection method was used to establish the stable transfected5-8F cells with TRIM29specific small interfering RNA (siRNA) vector and the stable transfected6-10B cells with TRIM29overexpression vector, and their corresponding control cell lines with empty vector transfection, respectively;(3) Western Blotting, MTT assay, flow cytometry,wound healing assay and Transwell were performed to compare the differences of protein expressional level, cell proliferation, cell cycle distribution, cell migration and cell invision in the stable transfected NPC cell lines. Using SPSS17.0statistical software for data analysis. Results:(1)The mRNA levels and the protein expressional levels of TRIM29in5-8F cells were significantly higher than in6-10B cells;(2) The stable transfected5-8F cells with TRIM29lower expression,6-10B cells with TRIM29overexpression and their corresponding control cell lines were established;(3) Compared with empty vector transfected and untransfected5-8F cells, the5-8F cells with TRIM29specific siRNA showed the decreased growth, increased percent of G0/G1phase cells, reduced percent of S phase cells, decreased migration ability and decreased invision ability. Compared with empty vector transfected and untransfected6-10B cells, the6-10B cells with TRIM29overexpression vector showed the increased growth, reduced percent of G0/G1phase cells, increased percent of S phase cells, increased migration ability and increased invision ability.Conclusions:(1) Overexpression of TRIM29plays an important role in promoting NPC cells growth, migration and invision;(2)TRIM29is closely related to the metastasis of NPC, may be the important molecular markers predicting metastasis of NPC, its specific mechanism and clinical application value remains to be further research.19Figures,7tables,63references...
