Objectives To observe the effects of the guanine nucleotide exchangefactor C3G overexpression on the survival of cardiomyocytes and itsunderlying mechanisms.Methods H9C2cardiomyocytes were transiently transfected withpCXN2-Flag and pCXN2-Flag-hC3G (overexpression of human C3GmRNA) plasmids, then treated with hypoxia/reoxygenation (H/R)respectively. The cardiomyocytes were divided into blank group, emptyplasmid group, C3G overexpression group, blank+H/R group, emptyplasmid+H/R group and C3G overexpression+H/R group. The expressionsof the C3G mRNA&protein and ERK1/2protein, and the apoptosis andproliferation rate were analyzed in the H9C2cardiomyocytes respectively.Results Compared with blank&empty plasimd groups and blank+H/R&empty plasimd+H/R groups, human C3G mRNA&protein, p-ERK1protein and proliferation rate were increased significantly, and apoptosis rate was decreased significantly in the C3G overexpression and C3Goverexpression+H/R groups respectively. Compared with blank+H/R andempty plasimd+H/R groups, p-ERK2protein was increased significantly inthe C3G overexpression+H/R group.(All P <0.05).Conclusions C3G overexpression can increase the ERK1/2proteinexpression, cell proliferation&survival and apoptosis inhibition in theH9C2cardiomyocytes; C3G overexpression also can decrease theH/R-induced inhibition of ERK1/2protein expression and cell proliferation&survival, and apoptosis in the H9C2cardiomyocytes. C3G overexpressioncan promote cell survival in the cardiomyocytes, it’s pro-survival effect maybe mediated by the increase of p-ERK1/2protein expression and apoptosisinhibition.
