Osteoporosis, an osteolytic bone disease, is a reduction in bone massdue to an imbalance between bone resorption and formation. In traditionalChinese medicine, Rhizoma Drynariae is commonly used in the treatmentof osteoporosis and bone nonunion. Modern pharmacological researcheshave indicated that naringin, which can induce the expression of theosteogenic marker in the osteoblast cell line and inhibition osteoclastsformation, is the main effective component of Rhizoma Drynariae.However, the specific mechanism is still not completely clear.Osteoprotegerin (OPG) has been identified as an important role inosteoblastic differentiation and bone formation. Therefore, componentsinvolved in OPG activation are good targets for the development ofantiosteoporosis drugs. In our study, we used RT-PCR and ELISA assaythat proved that naringin could enhance the effect of1α,25-dihydroxyvitamin D3(VD3) to promote the secretion of OPG in cellculture though increasing genes expression and protein levels. In addition, naringin can also effect the action of osteoclasts and bone lossing duringthe bone organ culture. This study identifies naringin as an effecter forosteoclast formation and bone resorption. Thus it makes a contribution tothe exploration of osteogenic differentiation agents in Chinese herbs andprovides evidence that natural compounds such as naringin might be analternative medicine for the prevention and treatment of osteolysis.
