Objective:To evaluate the influence of HRG on the human liver cancer and the mechanism.Methods:(1)Human liver cancer cells SMMC-7721were cultured in vitro with different concentration of HRG(25、50、100μg·ml-1).Cell growth inhibiting rate was detected by MTT assay, cell growth curve was detected by cell count assay, cell colony-forming rate and cell colony-forming inhibiting rate were detected by cell colony assay, cell cycle distribution were analyzed by flow cytometry assay.(2)Cell Migration and invasion were detected by scratch assay、transwell assay;the effects of HRG on the expression of Migration-related proteins(αvβ3and MMP-2) were detected by Western blot assay.(3)HepG2-CAM was treated with HRG, observing tumor volume, the number of blood vessels, tumor weight and inhibiting tumor rate.Results:(1) Human liver cancer cells SMMC-7721were treated with different concentration of HRG (25、50、100μg·ml-1):Cell morphology in different groups compared with the control group, there were varying degrees of change; the different concentration group of HRG inhibited the proliferation of tumor cells, the effects were a dose-dependent and time-dependent; The cell colony assay showed cell colony formation rate reduced, there were significantly difference between HRG and control group; The effect on cell cycle showed that HRG could change the cell cycle, there were significant blocking effect on tumor cells in S phase, entering a period of fragmentation of cell reduction, there were significantly difference between HRG and control group.(2)50μg·ml-1、100μg·ml-1HRG can significantly reduced cell migration; HRG could obviously inhibit cell invasion, there were significantly difference between HRG and control group. The expression of Migration-related proteins (avβ3and MMP-2) were reduced by HRG.(3) HRG has activities of inhibiting the growth of HepG2-CAM. Compared with the model control group,25μg·ml-1group P<0.05,50μg·ml-1group P<0.01,100μg·ml-1group P<0.001 Conclusion:(1) HRG inhibited the proliferation of Human liver cancer cells SMMC-7721and has a certain dose and time dependent as well as regulating cell cycle thereby inhibiting cell proliferation.(2) HRG has the ability to inhibit human liver cancer cells SMMC-7721migration and invasion, the mechanism of which may be related with reducing the expression of Migration-related proteins (avP3and MMP-2).(3) HRG has the ability to inhibit the growth of HepG2-CAM...
