Objective:To research the effect GSK-3β inhibitors lithium chloride on apoptosis andmechanisms of vivo rat myocardial ischemia-reperfusion injury. The study can offerclinical myocardial protection strategy for cardiopulmonary bypassMetholds:Selecting30clean male Sprague-Dawley rats, weight450-500g, were randomlydivided into three groups, group A for the normal control group (n=10), group B forextracorporeal circulation ischemia-reperfusion group (n=10),group C ischemiareperfusion and lithium chloride group (n=10). Three groups of rats has theanesthesia endotracheal intubation, mechanical ventilation, groups B and C linecardiopulmonary bypass intubation, cardiopulmonary bypass,Rat blood prefilleddeep hypothermic circulatory arrest (Deep hypothermic circulatory arrest, DHCA)myocardial ischemia-reperfusion model, to be recycled and stability, the rectaltemperature fell to16°C to18°C after about30min of cooling cardiac jump, stopmechanical ventilation give DHCA30min, then rewarming, and recovery ofmechanical ventilation, about50min rewarming to36.6℃, breathing on his own, tobe recycled and stability, stop the pump, disconnect the tubes, two hours after the ratswere sacrificed to stay take heart specimens.Immunohistochemical methods to detect negative myocardial cytoplasmicapoptotic factors Bcl-2and cytochrome C expression by RT-PCR assay themyocardial cells Caspase-3,8,9mRNA expression.Results:1. Negative apoptotic factor Bcl-2expression levels: Light microscopy showedthat Bcl-2positive cells stained pale yellow. Bcl-2expression in group B compared toA rat myocardium tissue expression was significantly higher (P <0.05). Than group B,group C, Bcl-2expression level was significantly increased (P <0.01). A, C There wasno difference (P>0.05).2. Cyt-C release level: normal myocardial cell cytoplasm rendered the scattered punctate Cyt-C immune reaction positive particles, Cyt-C release cytoplasmsignificantly increased in group B, and presented a sheet stained. The two groupscompared to the A, C expression levels significantly increased (P <0.05), A and C wasno difference (P>0.05).3. The expression levels of expression of Caspase family: Caspase3,8,9mRNAexpression in group A compared to group C was no significant difference (P>0.05), Bcompared to A, C two sets of expression levels were significantly higher (P <0.05).Conclusions:1, The preparation of the experimental application of rat cardiopulmonarybypass model for the study of myocardial ischemia-reperfusion injury modelexperimental reference.2, GSK-3β inhibitor lithium chloride, can be adjusted cytochrome C, Bcl-2expression of apoptosis-related factors, reduce apoptosis, has a protective effect onmyocardial ischemia-reperfusion injury.
