The medicinalmaterial is primarily applied to treate rheumatic diseases, stomachache, menalgia,tormina postpartum, traumatic injury, osteoarticular tuberculosis, medullitis, plotrickets in children, hepatitis, cirrhosis of the liver, etc. The existing standards ofBerchemia floribunda Brongn. has been contained in The Jiangxi Medicines StandardEdition1996page176, which determination of content is vacancies and only has therational identification, which has no TLC identification, the standard has notcomplete, it is hard to control the quality of medicinal materials and their preparations.So raise the standard be imperative.In order to study the larger polar water-soluble compoments of Berchemiafloribunda Brongn., our group first study the extraction, separation and structureelucidation of chemical constituents of Berchemia floribunda Brongn.. Our groupisolated from this plant four compounds, we identified as Aromadendrin4’-O-β-D-glucopyranoside、Rubrofusarin6-gentiobioside、4-hydroxy benzoic acidand3,4-two hydroxy benzoic acid, and they all having larger polar. Which for the firsttime in the Berchemia floribunda Brongn. herbs found Rubrofusarin6-gentiobiosidefor Naphthopyran ketone glycosides,the content determination has not yet beenreported at home and abroad, and it has high content in Berchemia floribundaBrongn., According to the literature Rubrofusarin6-gentiobioside vitro antioxidantactivity, also has hepatoprotective activity[17-19], it can representative for theBerchemia floribunda Brongn. quality control.Our group used HPLC method for simultaneous determination of Aromadendrin4’-O-β-D-glucopyranoside and Rubrofusarin6-gentiobioside content. The method was using ODS column with acetonitrile(A)-0.2%phosphoric acid(B) as mobile phase,and gradient elution(0~10min,18%A;10~12min,18%A→23%A;12~35min,23%A;35~38min,23%A→18%A;38~60min,18%A). The flow rate was1.0ml﹒min-1and the detection wavelength was277nm. The column temperature was30℃.Results:Aromadendrin4’-O-β-D-glucopyranoside in the range of0.03006~0.6012μgmass and peak area was a good linear relationship, the standard curve equation was Y=1592346.1281X+34255.1880,correlation coefficient R2=0.9943(n=5),the averagerecovery was97.73%, RSD:1.05%.Rubrofusarin6-gentiobioside in the range of0.0306~0.612μg mass and peakarea was a good linear relationship, the standard curve equation was Y=3323971.1546X+63706.3648,correlation coefficient R2=0.9954(n=5),the averagerecovery was97.62%, RSD:1.38%,with good reproducibility.
