| Radix Adenophorae is the dried root of Adenophora tetraphylla (Thunb.) Fisch. and Adenophora stricta Miq. It has been recorded in Chinese Phamacopocia all the time. For commonly used blindly Yin drugs, it can not only nourish Yin and clear lung, but also benefit stomach, reduce phlegm and tonify Qi. Radix Adenophorae commonly used in hot dry cough, lung Yin deficiency fatigue syndromes, glue dry cough phlegm, stomach Yin deficiency, eat less vomiting, Qi and Yin deficiency, vexed hot dry mouth, etc. Generally containing polysaccharide in radix adenophorae, terpenoids, phenolic acids, coumarin, sterols and volatile oil chemical composition, Radix Adenophorae has a cough expectorant, anti-oxidation, anti-aging, immunity, resist radiation and other pharmacological effects. Radix adenophorae is widely used, but its research reports on chemical composition, material base and quality control are lesser.In order to detect the therapeutic material basis of Radix adenophorae, control the quality of medicinal materials, and ensure the safety of medicine usage, this study analyzed its chemical composition and devised its quality control method.The contents include:1 Studies on Chemical Constituents of Radix adenophoraeTo study the chemical constituents of Adenophora tetraphylla. The compounds were isolated and purified by silica gel column, sephadex LH-20 column, Cs column, C18 column and HPLC chromatography. Their structures were identified by physicochemical properties and spectral data-UV,1H-NMR,13C-NMR, ESI-MS. Sixteen compounds have been isolated from the Petroleum ether, EtoAc and n-butanol fraction. They were respectively identified as a-amyrin acetate (1), taraxerone (2), lupenone (3), β-sitosterol (4), lupeol acetate (5), sessilifolic acid 3-O-isovalerate (6), stigmastane (7), n-pentadecanoic acid (8), bis(2-ethylhexyl) phthalatel (9), wogonoside (10), guaiacol β-D-glucopyranoside (11),5-hydroxymethyl-furfural (12), baicalin (13), Stearic acid a-propyl ester (14), lp-hydroxybenzoic acid glucoside (15), 0-Octadecenoic acid (16).Among them compound 1,8,10,11,13 were first obtained from the Campanulaceae, and compound 9,12 were obtained from the Adenophorae for the first time, and compound 6,14,15,16 were obtained from this plant firstly.2 Study on the measurement methods for the contents of Radix adenophoraeAn HPLC method was established for simultaneous determination of three triterpenoids (taraxerone, lupeol acetate, sessilifolic acid 3-Oisovalerate) in Radix adenophorae. Simple and sensitive, this method was applied to determing the contents of 11 batches of Radix adenophorae.3 Study on the HPLC Fingerprint of Radix adenophoraeThis research establish a preliminary HPLC fingerprint of Radix adenophorae, and employed the "TCM Fingerprint Similarity Evaluation Software(version 2004)",which is developed by Chinese Pharmacopoeia Commission, in evaluating the similarity of 11 batches of Radix adenophorae. It is found that a tatal of 14 characteristic common peaks were identified, with the similarities ranging from 0.843 to 0.958. |
