Study On The Pharmacy Of Zhi Yu Granules

Objective:To determine the antidepressant part of zhi yu capsule and optimize theoptimum extraction process，then to prepare the granule with the extracts，establishthe quality standard of zhi yu capsule and finally carry on a preliminarily discussionof the antidepressive mechanisms of zhi yu capsule．Method: With the accumulation immobility time as a index， to screen theantidepressant part of zhi yu capsule by forced swimming test；to determine thecontent of geniposide with HPLC and the curcumenol with UV-Vis，with the contentof geniposide and curcumenol as indexes，the extraction process of liposolubleconstituents in zhi yu capsule was optimized by L9（34）orthogonal experiment；todetermine the content of total polysaccharides with UV-Vis and with the content as aindex，the extraction process of water-soluble constituents was optimized by L9（34）orthogonal experiment； With the formability and dissolubility of granules asindexes，to select wetter、the kind and amount of accessories and the forming processusing dry granulating technology by single factor experiment；to establish the qualitystandard of zhi yu capsule by the regulation of “granules general principles” inChinese Pharmacopoeia(2010Edition)Volumn I；to determine the optimum dose byanimal test and establish distribution in vivo of the main active componentpillsgeniposide of zhi yu capsule．Result: The extracts of zhi yu prepared by pulsing the amount of60%ethanolsolution of herbs and plusing the amount of water of herbs，extracting two times hadthe best effect of antidepressant；the best extraction technology of zhi yu was pulsingthe amount of60%ethanol solution for ten times of herbs，extracting two times，eachtime1.5hours and plusing the amount of water for ten times of herbs，extracting threetimes，each time1.0hours；the animal test indicated that the pesticide effect of zhi yucapsule was stable，the distribution law of the main active component pillsgeniposidewas:liver＞kidney＞lungs＞spleen＞heart＞brain； The spots of FructusGardeniae、Radix Curcumae and Coptis Chinensis on TLC plates were clear withoutinterference in the blank reference；the average content of geniposide and total polysaccharides were2.4％and6.0％，zhi yu capsule accord with the regulation of“granules general principles” in Chinese Pharmacopoeia(2010Edition)Volumn I．Conclusion: The extraction process and preparation technology of zhi yu capsulewere safe and reliable with good reproducibility， and the pesticide effect was stable；the methods of TLC were simple，feasible and repeatable with stable and accurateresult，quality inspections accord with relative requirement．...
