ObjectiveTo observe the effects of estradiol (E2) and progesterone (P) on theexpression of FGF1, FGFR1, MMP2,MMP9, and on the proliferation, invasionand migration of the MA891breast cancer cells, and preliminarily analysethe mechanism of invasion and migration of MA891breast cancer cells.Methods1. Abstract the tumor tissue from TA2tumor-bearing mice, primary culturethe spontaneous breast cancer cells(MA891breast cancer cells)of theTA2mouse. Inoculating them in the TA2mice subcutaneously after stablepassage, and then observe the growth of the tumor.2. To observe the effects of E2or P alone and combination of E2and P onthe invasion and migration of MA891breast cancer cell by using theTranswell and the wound-healing assay3. The effects of E2or P alone and combination of E2and P on the expressionof FGF1, FGFR1,MMP2,MMP9were determined by cell immunohistochemistryanalyse.4. The optical density (OD) value representing the expression of FGF1,FGFR1, MMP2, and MMP9was measured using Image-Pro Plus6.0. Statisticalanalysis was performed with SPSS13.0software.. The values werecompared by one-way analysis of variance, followed by post hoc analysisby the least-significant difference test. P <0.05was consideredsignificant.Results1. Ten mice inoculated subcutaneously with MA891breast cancer cells allhad tumor growth in18days 2. The effects of E2or P alone and combination of E2and P on the invasionand migration of MA891breast cancer cell. Comparing to the control group,Transwell invasion and migration test shown that E2, P, E2+P treatmentsignificantly increased the invasion and migration of MA891breastcancer cells (P<0.05), and the ability of invasion and migration ofMA891breast cancer cells after E2+P treatment was higher than E2or Ptreatment alone.3. The effects of E2or P alone and combination of E2and P on the expressionof FGF1, FGFR1,MMP2,MMP9of MA891breast cancer cells. Cellimmunohistochemistry analysis shown that E2and P treatment couldupregulate the expression of FGF1, FGFR1, MMP2, MMP9of MA891breastcancer cells (P<0.05,P<0.05,P<0.05,P<0.05), and the expressionof FGF1, FGFR1, MMP2,MMP9was higher after E2+P treatment than E2orP treatment alone.(P<0.05,P<0.05)Conclusion.1. E2and P have positive regulation on proliferation, migration andinvasion of MA891cells.2. The increased expression of FGF1, FGFR1, MMP2,MMP9of MA891cells, andthe increased activity of MMP2, MMP9might increased migration andinvasion of the cells.
