Objective:By the modifided Allen’s weight drop method,a model of spine cord injury in rats was maken. By the studying the effect of teramethylpyrazine(TMP) on the expression of GAP-43and Survivin,the possible mechanism of TMP on the recovery of motor function after spinal cord injury was explored.Methods:72adult Sprague Dawley rats (male or female)with250-270g were randomly divided into three groups:sham operation group (group S, n=12)normal saline control group (group N,n=30)、 tetramethylpyrazine treatment groupT,n=30).Every group was further divided into6sub-groups(group S,n=2.group N and group T,n=5). Each sub-group rats were killed at1,3,7,14,21and28days postoperativly. The models of spine cord injury in rats at T10level were maken by the modified Allen’s weight drop method in group N and group T.But group S only received the laminectemy.About30minutes after the surgery, a dose of200mg/Kg body weight of TMP was injected via peritoneal, for group T,once a day for5days.and the group N rats were injected with the same dose of saline solution(NS),Group S rats didn’t be injected.Then the motor function of three gruop rats were evaluated with BBB score at each time point.The injury spinal cord tissue was received at the each time point by killing the rats.The tissue was cut into10μm thickness section and used to evaluate the expression of GAP-43or Survivin with immunohistochemistry methods. The software SPSS16.0was used to analyze the results with statistical methods.Results:1.BBB scores:At7d,14d,21d and28d,the BBB scores of group T rants were higher than those of group N rats(P<0.05).2.The expression of GAP-43:The positive cells with GAP-43positive expression were not founded in group S rats.The positive cells with GAP-43positive expression were up to the maximum in group T and in group N at14d.At3d,7d,14d,21d, the positive cells with GAP-43positive expression in group T were more than those in group N (P<0.05).3.The expression of Survivin:The positive cells with Survivin positive expression were not founded in group S rats yet. At1d,3d,7d,14d, the positive cells with Survivin positive expression in group T were more than those in group N (P<0.05),The positive cells with Survivin positive expression were up to the maximum in group T and in group N at7d.Conclusions:1.TMP has positive effect on the recovery of motor function after spinal cord injury.2.TMP can improve the expression of GAP-43and Survivin in cells after spine cord injury.
