Objectives:To study the effects of hydrogen sulfide (H2S) the pathological changes, the expression of interleukin-4(IL-4) and macrophage inflammatory protein-2(MIP-2) in ALI rats, and determine the possible protection role of hydrogen sulfide in LPS-induced ALI rats.Methods:27healthy male SD rats were randomly divided into normal control group, ALI group and NaHS intervention group, n=9.(1) normal control group:intraperitoneal injection of saline0.5ml,10minutes after intravenous injection of saline (2ml/kg).(2) ALI group: intraperitoneal injection of saline0.5ml,10minutes after intravenous injection of LPS (5mg/kg) manufacturing rat model of ALI.(3) NaHS intervention group:intraperitoneal injection of NaHS (28μmol/kg),10minutes after intravenous injection of LPS (5mg/kg). The general state changes of the rats was observed in each group, including the spirit of the situation, cyanosis and so on; six hours after the intravenous injection,all the rats were sacrificed, specimens from the BALF and lung tissue of the rats were collected correspondingly. Visually observed lung tissue generally changes, Take the right upper lung with formalin-fixed, paraffin-embedded sections of HE staining, light microscope to observe the pathological changes of the lung tissue; Anatomic neck exposed tracheal intubation, and lavaged, Each time the4℃saline solution2mL repeated lavage3times, and filtered with a single layer of gauze, centrifugation underwent total cell,count cell counts were observed by the total number of cells in BALF, the concentrations of IL-4and MIP-2in BALF were measured by the enzyme joins immunosorbent assay (ELISA).Results:1. Changes in the total number of lavage fluid cells:the total number of cells in BALF of normal control rats was2.16±0.36(×109/L); Ali rats BALF total number of cells to13.52±2.73(×109/L) between the two groups ALI rats BALF total number of cells was significantly higher (P<0.05); NaHS intervention group BALF total number of cells9.92±1.93(×109/L), lower than the ALI group, but still higher than the normal control group, the difference statistically significant (P<0.05)2. Lavage fluid IL-4changes:normal control group rat BALF IL-4concentration of5.28±0.32pg/ml; the ALI group BALF IL-4concentration is5.99±0.63pg/ml, with the normal control group increased significantly (P<0.05); NaHS intervention group BALF IL-4to6.62±0.59pg/ml, than the ALI group increased, and the difference was statistically significant (P<0.05)3. Lavage fluid MIP-2changes:normal control group rats in BALF MIP-2concentration is1.66±0.18pg/ml the ALI group in BALF MIP-2concentration of2.99±0.79pg/ml, normal control group significantly increased (P<0.01); NaHS intervention group BALF MIP-2concentration of2.14±0.19pg/ml than the ALI group reduced, but still higher than normal controls, and the difference was statistically significant (P<0.05)Conclusion:1.The expressions of IL-4and MIP-2in BALF are increased.2. Hydrogen sulfide could promote the expression of IL-4, while inhibit the expression of MIP-2in ALI.3.Hydrogen sulfide may have a protective effect on LPS-induced ALI.
