Objective Combining EMT and CSCs theories, this study preliminarily researched on the difference of proliferation and the expression of biomarkers related to epithelial-mesenchymal transition and cancer stem cell in cell line ACC-M induced by TGF-β1and discusses its significanc.Methods (1) ACC-M cells containing EdU were observed under fluorescence microscope before and after induced by TGF-β1and paired t test was used for statistical analysis.(2) The expression of EMT related markers E-cad, N-cad, Vimentin, MMP-2and CSCs related markers Bmi-1, Oct-4, CD44and CD24in ACC-M cells after induced by TGF-β1on Oh,12h,24h,48h and72h were detected with Western blot technology and the correlation among them was analyzed.Results (1) EdU positive ACC-M cells obviously increased afte induced by TGF-β1, P<0.05.(2) The expression of E-cad, Oct-4and CD24decreased while the expression of N-Cad, Vimentin, MMP-2, CD44and Bmi-1increased in ACC-M cells after induced by 10ng/ml TGF-β1and there were linear correlations among EMT and CSCs related markers, P=0.000<0.05.Conclusion (1) TGF-β1can promote proliferation of ACC-M cells obviously;(2) TGF-β1can induce EMT in ACC-M cells, accompanied by the changes of CSCs marker expression changes.
