Nanobiotechnology is widely studied all over the world. This field involves various knowledge in physics, chemistry, medicine, materials science, biology, and other branches. Based on the research achievements in these fields, nano-biotechnology has addressed a significant development in recent years. Nanometer biological material reflects directly the developing level of nano-biotechnology. One of the most important biological nanomaterials is smart drug carrier., and this field has been high developed since the80’s in20th century. To ensure the safety, nano drug carriers should pass the cellular experiments in vitro before used in a living organism.Recently, quantum dots as fluorescent probes in the application of biological analysis has been a hot research topic. Excellent photoluminscent property and low toxicity make zinc oxide as a promising candidate as fluorescent probes. The present difficulties mainly lie in the synthesis method.Based on the status mentioned above, we did the following work:First, we synthized a type of hollow nano carriers using chitosan, acrylic acid and β-cyclodextrin. The nanoparticles were characterized and loaded with anti-cancer drug PTX. C6glioma cells were cultured for24h prior to the assay and then incubated with PTX, PTX-loaded nanoparticles and blank nanoparticles at different concentrations. The cytotoxicity of the polymer and the cytotoxicity against C6glioma cells between bare PTX and PTX-loaded nanoparticles were studied by MTT assay. We Synthesized FITC-labeled nanoparticles and stuidied the cellular uptake of nanoparticles using flow cytometry and laser confocal fluorescence microscopy. The result declared the non-toxic of this polymer. It also showed that the polymer had a controlled release effect and the number of particles entered the cells increased with time.We also studied a new method for preparing ZnO quantum dots. It’s a one-step preparation involving only two reagents-zinc acetate and ethanol in a mild reaction condition. Three ways of synthesizing ZnO quantum dots were introduced. Besides this new method, were two widely used methods invented by Spanhel and Oskam respectively. The products were studied using TEM, HRTEM, XRD, XPS, UV-Vis Spectrum and PL Spectrum. According to the result, this new method is fairly easy and the products possess excellent PL properties and high stability at room temperature.
