ObjectiveIn order to research the relationship IL-17, IL-23and acute graft-versus-host disease (aGVHD) in clinical allogeneic bone marrow transplantation (allo-BMT) recipients, we conducted a prospective analysis of peripheral blood IL-17,IL-23, which may have important prognostic value as a biomarker of acuteGVHD.MethodsThe level of IL-17, IL-23in the plasma were got by Enzyme Linked Immu-nosorbent Assay (ELISA). The relative expression of IL-17mRNA and IL-23mRNA transcripts in PBMCs of allo-HSCT patients was detected by real timepolymerase chain reaction (Realtime-PCR).Results(1)The test result of IL-17, IL-23in plasma in each group: comparedwith normal control group, the concentration of IL-17, IL-23in aGVHD po-sitive group was significantly increased,the difference was statistically sign- ificant (p=0.007, p=0.005),the concentration of IL-17, IL-23in negative gro-up was no evident difference, there was no statistically significant differe-nce(p=0.67, p=0.87); Compared with aGVHD negative group, IL-17, IL-23concentration in aGVHD positive group was significantly increased, thedifference was statistically significant(p=0.008, p=0.002);(2)The test result of IL-17, IL-23concentration in plasma in aGVHDpositive group at different time points: compared with14days before the aGVHD occurrence, IL-17, IL-23concentration in the plasma in aGVH-D positive group occurred on the same day was significantly increased,t-he difference was statistically significant (p=0.046, p=0.033); comparedWi-th aGVHD occurred after7days,14days,21days, IL-17, IL-23conc-entration in the plasma in aGVHD occurred on the same day was signi-ficantly increased, the difference was statistically significant (p=0.045, p=0.021, p=0.010). Concentration of IL-17, IL-23peaked, when aGVH-D occurrence. with symptom control, gradually decline;(3)The test result of IL-17mRNA, IL-23mRNA relative expression inperipheral blood mononuclear cells: compared with control group, the relativeexpression of IL-17mRNA, IL-23mRNA in aGVHD positive group wassignificantly increased, the difference have statistical significance (p=0.042, p=0.015), and the relative expression of IL-17, IL-23mRNA in aGVHD negativepatients has no obvious change, there was no statistically significant difference(p=0.78, p=0.82). Compared with aGVHD negative patients,the relativeexpression of IL-17, IL-23in aGVHD positive group increased significantly,the difference was statistically significant (p=0.021, p=0.017).ConclusionThe level of IL-17and IL-23were all increased in the patients with aGVHDwhich may have important prognostic value as a biomarker of aGVHD.
