ObjectiveTo explore global ischemic injury in hippocampal CA1region of rat brainPARP-1and Caspase-3expression.MethodsRandomly select healthy adult SD rats,Global ischemia rats by adopting themethod of4-VO model. Intraperitoneal injection of10%chloral hydrate (3mL/kg) were anesthetized, and exposed to double flank holes separated aftercoagulation of the vertebral artery blockage, Two days again after anesthesia,isolated bilateral carotid artery occlusion using intact carotid artery clip causingglobal ischemia,0,5,10,15,20,25,30min ischemia time respectively time point (n=14) for coverage. TTC staining using rat cerebral ischemic volume of theregion, calculate the volume of brain tissue infarction accounted for thepercentage of brain volume. Using optical microscopy and electron microscopyglobal ischemia in rats hippocampal CA1region of the brain morphologicalchanges; respectively by immunohistochemistry and Western blot were used todetect rat hippocampal CA1region poly adenosine diphosphate ribosepolymerase-1(PARP-1) and cysteine proteases-3(Caspase-3) proteinexpression.ResultsThe positive expression of PARP-1is located nerve nucleus in thehippocampal CA1and the positive expression of active-caspase localized in thecytoplasm.PARP-1showed a gradually rising trend in different ischemic timepoint, then keeping stable after the peak has stabilized. While the active-caspase show lower expression levels.ConclusionsGlobal ischemia30min memory in neuronal apoptosis and oncosis. Which,PARP-1activation leads to excessive Oncosis, and with the extension ofischemic time increased expression; Caspase-3activation can lead to apoptosis,expression was low early in the ischemic state.
