| ObjectiveStudy on different expression correlation between dampness-heat of spleen-stomach and liver depression and spleen deficiency of chronic hepatitis B to explore the microRN A expression profiles and nature of chronic hepatitis B with dampness-heat of spleen-stomach and liver depression and spleen deficiency,provide a objective basis of differentiation in clinic.MethodsApplying gene chip technology to test the chronic hepatitis B patients with syndrome of liver depression and spleen deficiency and spleen-stomach dampess-heat,observe the differential expression between the syndromes.All date were analyzed with the spss19.0software.meanwhile using SAS system to select the microRNA and functional analvsis.Results1Compare with control group,98differently expressed microRNAs were found in dampness-heat of stomach and spleen group,64microRNAs were up regulation,34microRNAs were down regulation.There are statistically significant differences in the77microRNAs (P<0.05),60microRNAs were up regulation (P<0.05),17microRNAs were down regulation (P<0.05).Meanwhile predict the target gene of77microRNAs,involved to3104target genes which1755target genes were up regulated,1349target genes were down regulated.2Compare with control group,72differently expressed microRNAs were found liver depression and spleen deficiency group,45microRNAs were up regulation,27microRNAs were down regulation.There are statistically significant differences in the41microRNAs (P<0.05),32microRNAs were up regulation (P<0.05),9microRNAs were down regulation (P<0.05).Meanwhile predict the target gene of41microRNAs,involved to1385target genes which900target genes were up regulated,485target genes were down regulated.3The liver depression and spleen deficiency group compare with dampness-heat of stomach and spleen group,6differently expressed microRNAs were found (P<0.05) which1microRNA were up regulation,5microRNAs were down regulation Meanwhile predict the target gene of them,involved to1125target genes which274target genes were up regulated,851target genes were down regulated.Conclusions1The differential expression of microRNA(spectrum) between chronic hepatitis B with syndrome of spleen-stomach dampness-heat and liver depression and spleen deficiency are:hsa-miR-1273g-3p, hsa-miR-197-5p, hsa-miR-320d, hsa-miR-4534, hsa-miR-5703, hsa-miR-642b-3p.The differential expression of microRNA may related to some biological basis such as:process of actin filamentã€hormone metabolism etc.2Compare with the normal persons,the level of microRNA in the chronic hepatitis B with syndrome of spleen-stomach dampness-heat group had significantly difference.
