Compared with others’ grape,spine grape is a fruit of berry which has more seeds, smaller granule and higher amount of procyanidin. So it will be of importance to do research of spine grape seeds procyanidins’ separation&purification technology and function evaluation in the food and pharmaceutical industry. The researches have shown that procyanidin is a kind of good antioxidant, free radicals scavenginger which has many functions such as anti-tumor, Anti-mutagenicanti-radiation, anti-viral, anti-fungal,and can alleviate cardiovascular disease. Anti-aging research of procyanidin is a hot spot, the purpose of this paper is to research the separation&purification technology of spine grape seeds procyanidin and evaluation of anti-aging function.1.At first after separation&purification of spine grape seeds procyanidin, make it sure that the procyanidin concentration is more than95%. It take measures of supercritical CO2extraction and macroporous resin adsorption to extract procyanidin from spine grape in this paper. The process is as following:procyanidin was extracted form grape seed oilby the way of supercritical CO2extraction technology.The extracted residue was merged and concentrated by the aqueous alcohol solution.Then after alcohol-precipitation, decompression, evaporation and dryness, the procyanidin can be adsorbed by the asthenic-polar macroporous resin AB-8.After desorption and evaporation, the product can be collected.This method is of high precision.After several-times adsorption, concentration of proanthocyanidins can reach96.38%.2. By UV-B irradiation, cultivated mouse fibroblasts L929was established to be the cell radiation damage model. After incubation with the different doses of procyanidin extracts, it showed that ultraviolet radiation deteriorate injury of L929cell by electron microscopy and flow cytometry analysis. It is significant protective effect for procyanidin-pretreated cells model can effectively resist ultraviolet radiation damage。3. In this paper,it research the measurement methods and functional evaluations of spine grape seeds procyanidin in scavenging the four common free radicals, the results showed that the procyanidin of spine grape seeds has strong functions of free radical scavenging and antioxidant resistance. 4.By establishing HSA/MDA reaction system, the anti-aging function of procyanidin was evaluated. By comparing between the differance after and before HAS/MD A reaction system, it showed that procyanidin can prevent APFs to generate actively and had a good inbibitory function against APFs generation...
