Object:Nasopharyngeal carcinoma is commom type of head and neck tumors, lymphnode metastasis can be occurred early stage of disease. Radiation therapy is the firstchoice for the treatment of nasopharyngeal carcinoma. Followed with the updatingof radiotherapy equipment and technology, the overall5-year survival rate fornasopharyngeal carcinoma can reach60%to70%.The local recurrence and distantmetastasis is the main reason for failure of nasopharyngeal cancer treatment.Metastasis is the characterization of malignancies. It is the result of comprehensiveinteraction of multi-factor, multi-step and multiple-gene. In recent years, tumormetastasis suppressor genes become a hot topic. KISS1/hot7T175, KAI1/CD82,BRMS1, TIP30/CC3, Maspin, MKK4, MTSS1is newly developed tumor metastasissuppressor gene. In this study, the expression of these seven genes was investigated innasopharyngeal carcinoma tissues, and the relation to lymph node metastasis anddistant metastasis were analyzed. Further, the genes function in nasopharyngealcarcinoma metastasis were explored.Method: The protein level of KISS1/hot7T175, KAI1/CD82, BRMS1, TIP30/CC3,Maspin, MKK4and MTSS1were determined in100cases of nasopharyngealcarcinoma by immunohistochemistry SP method. And, the relationship of lymph nodemetastasis and distant metastasis of nasopharyngeal carcinoma were also analyzed. Result:1, Lymph node metastasis of nasopharyngeal carcinoma and KISS1/hot7T175,KAI1/CD82, Maspin protein expression was negatively correlated (P=0.006; P=0.035; P=0.02; P <0.05, statistically significant); and MTSS1protein expressionwas positively correlation (P=0.005; P <0.05, statistically significant); andTIP30/CC3, BRMS1, MKK4protein unrelated (P>0.05, not statistically significant)2, Protein expression of distant metastasis of nasopharyngeal carcinoma with KISS1negative correlation (P=0.014; P <0.05, statistically significant); MTSS1proteinexpression was positively correlated (P=0.002; P <0.05, statistically significance);and KAI1/CD82, BRMS1, TIP30/CC3, Maspin and MKK4protein unrelated (P>0.05,not statistically significant)Conclusion:1,KISS1/hot7T175protein in nasopharyngeal carcinoma and distant lymph nodemetastasis in both the loss of expression of the gene may inhibit metastasis ofnasopharyngeal carcinoma and lymph node metastasis.2,KAI1/CD82, Maspin protein in nasopharyngeal carcinoma lymph node metastasisin both the loss of expression of these two genes may inhibit lymph node metastasisof nasopharyngeal carcinoma.3,MTSS1proteins were overexpressed in lymph node metastasis and distantmetastasis of nasopharyngeal carcinoma, nasopharyngeal cancer gene may promotelymph node metastasis and distant metastasis.
