【Objective】: Nasopharyngeal carcinoma is a common malignant tumor inotorhinolaryngology. Radiotherapy is the first choice for treatment of nasopharyngealcarcinoma because of the hidden position of nasopharyngeal and complex anatomicalstructures around.With the continuous advancement of radiotherapy techniques, theaverage of5-year survival rate is only about60%-70%in nasopharyngeal carcinoma.Tumor distant metastasis is the major reason according to recent informations,itaccounts for about56.2%.Tumor metastasis is a complex process, and is controlled bypromoting the transfer genes (PTMG) and metastasis suppressor genes (MSG). Studiesshow there are lots of malignant tumors with Kai-1/cd82gene’s abnormalexpression.This new metastasis suppressor gene plays an important role in the processof tumor metastasis. Based on above background, we detected kai-1/cd82mRNA andkai-1/cd82protein expression levels in tissues of normal nasopharyngeal andNasopharyngeal Carcinoma.by methods of Immunohistochemistry and RT-PCR, weinvestigated the relationship between abnormal expression of kai-1/cd82gene andfactors related, such as: age、 gender、pathological type、T stage and lymph nodestaging in order to improve curative effect and lower metastasis incidence ofNasopharyngeal Carcinoma,also would open up new ways for its prognosis and makean useful attempt to gene therapy in the future.【Method】:1. Cd82expression level were detected in70cases of nasopharyngeal carcinoma and30cases of Non-tumor nasopharyngeal tissues by immunohistochemistry.2. The kai-1/cd82mRNA expression level were detected in28cases ofnasopharyngeal carcinoma and15cases of Non-tumor nasopharyngeal tissues byreverse transcription polymerase chain reaction (RT-PCR).【Results】:1. The positive expression rate of kai-1/cd82mrna in nasopharyngeal carcinoma is42.9%(12/28), is lower than73.3%(11/15) in non-tumor nasopharyngeal tissues,the difference has no statistically significant (p=0.056). 2. The positive expression rate of kai-1/cd82protein in nasopharyngeal carcinoma is44.3%(31/70), is lower than70.0%(21/30) in non-tumor nasopharyngeal tissues,the difference has statistically significant(p <0.05).3. There is no correlation with kai-1/cd82mrna expression level and the factors suchas,sex, age, pathologic type, T stage and other clinical parameters in nasopharyngealcarcinoma(p>0.05). In stage of N0,N1, N2, N3, the positive expression rate ofkai-1/cd82mrna is85.7%(6/7),55.6%(5/9),10%(1/10),0(0/2),it is graduallydecreased with progresses of the N stages.In the group without lymph nodemetastasis (N0) the rate is85.7%(6/7), is significantly higher than28.6%(6/21)in the group with lymph node metastasis (N1-3).the difference has statisticallysignificant (p <0.05).among the groups with Lymph node metastasis (N1, N2, N3),the differences between the each other groups is not significant (p>0.05).4. There is no correlation with kai-1/cd82protein expression level and the factors suchas, sex, age, pathologic type, T stage and other clinical parameters innasopharyngeal carcinoma(p>0.05). In stage of N0,N1, N2, N3, the positiveexpression rate of kai-1/cd82protein is68.4%(13/19)、43.8%(7/16)、33.3%(9/27)、25(2/8),it is gradually decreased with progresses of the N stages.In thegroup without lymph node metastasis (N0) the rate is68.4%(13/19), significantlyhigher than35.3%(18/51)in the group with lymph node metastasis (N1-3).thedifference has statistically significant (p <0.05).among the groups with Lymph nodemetastasis (N1, N2, N3), the differences between the each other groups is notsignificant (p>0.05).【Conclusion】:1. Kai-1/cd82gene expression level is low in nasopharyngeal carcinoma,while is highin non-tumor nasopharyngeal tissues, our conclusion suggests that kai-1/cd82genemay play an important role in the pathogenesis and the development ofnasopharyngeal carcinoma,.2. Low expression of kai-1/cd82gene is associated with lymph node metastasis ofnasopharyngeal.It suggests that kai-1/cd82gene might be one of the factors thatinhibit lymph node metastasis in nasopharyngeal carcinoma.
