| Objective: Take the study on the chemical consitituents from Onychium lucidumand the pharmacological activity of onychin. To provide basis for obtaininganti-cancer and anti-oxidation lead compounds with strong biological activity and lowtoxicity.Methods:(1) Isolate and analyze the effective constituents of Onychium lucidum byusing modern separation technology. Then we identified their structures by the meansof many modern organic spectral analysis, such as IR, MS,1H-NMR,13C-NMR etc.(2) The anti-cancer activity of onychin on MGC-803cell lines, HeLa cell lines andHepG2-A16cell lines was evaluated by the MTT-based assay. The anti-cancer effectof onychin on the survival time of mice inoculated with Ehrlich ascites carcinoma wasdetected in vivo.(3) Study the anti-oxidant activity and the mechanism of onychin onoxidative damage of vascular endothelial cells by animal experimental method andMTT-based assay.(4) Take the acute toxicity study of onychin on normal SD ratmodels by using animal experimental method.Results:(1) Sixteen compounds were isolated from Onychium lucidum, and thirteenof their structures were identified.(2) Onychin had anti-proliferaive effect andanti-colony forming ability on MGC-803cell lines, HeLa cell lines and HepG2-A16cell lines. Onychin could extended the survival time of mice inoculated with Ehrlichascites carcinoma.(3) The results of onychin’s in vitro antioxidant activity testshowed that onychin had a protective effect on vascular endothelial function whichwas damaged by oxidants.(4) The rats were given intraperitoneal injection ofOnychin of30mg·kg-1and150mg·kg-1each day for7days, no rats showed toxicitychange in the appearance, hemogram, liver and kidney function, a variety of organs, tissues and bone marrow.Conclusion:(1) Sixteen compounds were isolated from Onychium lucidum, andthirteen of their structures were identified;(2) Onychin possessed anti-tumor activityand antioxidant activity, and its toxicity was low. |
