Pain Capsule Anti-inflammatory And Analgesic Effects And Acute Toxicity Test Experimental Study

Objective:To study the effects of Anti-inflammatory and Analgesic of Chinese traditional medicine of Gutong capsule, and the reliability of oral administration of this medicine. Making the foundation of further discovering the mechanism of Gutong capsule, so provide a theoretic basis for the clinical application of Gutong capsule.Methods:1 Analgesia test1.1 Hot plate test: Assay the female mice' s background pain threshold on the 55 ± 0.5℃ hot plate. Choose 50 mouse according with the standard. Then divide the mouse equally into 5 groups: group of normal sodiume, group of Anti-Hyperosteogeny capsule,group of low dose,medium dose and high dose Gutong capsule. Intragastric administration on each mice with certain dose, once daily for seven days. After the last administration, assay the mice's pain threshold again at different time.1.2Writhing test:Choose 50 mouse, the group dividation and administration are the same as the experiement above. 30 minutes after the last administration, inject 0.2ml 0.5% glacial acetic acid into mice' s abdominal cavity. Record the times of wrenching their body in 15 minutes.2 Anti-inflammatory test2.1 Auricle swelling test:Choose 50 mouse, the group dividation and administration are the same as the experiement above, 30 minutes after the last administration, inunct 0.03ml dimethylbenzene on the right auricle. One hour later,kill all of the mouse, and cut circle auricle of every mice's left and right auricle. The difference of the circle auricle is the edema of the auricle.2.2Foot swelling test: Choose 50 rats, the group dividation is the same as the experiement above. Assay the right hindfoot's volum of each rat. 30 minutes after the last administration, inject 0.1ml 1% carrageenin subcutaneously in the middle of rat' s right hindfoot. Then assay the volum of each rat' s right hindfoot once per hour for six times. Record the results and calculate the edema inhibition ratio of the rat' s hindfoot.3 Acute toxicity test: Choose 40 mouse, and divide the mouse equally into 4 groups. Intragastric administration on each mice with different concentration water extract of Gutong capsule. Then observe the mice' s weight, skin, breath,activity, mental state, food and drink, urine and stool for seven days.Record the observation results.Results:1Analgesia test: Gutong capsule can significantly diminish pain caused by chemical(H~+) and physical (heat) stimulation.2 Anti-inflammatory test: Gutong capsule can significantly inhibit rat paw edema and mice auricle edema.
