ObjectiveThrough electroacupuncture stimulation on Zusanli acupoint and Ashixue acupoint in Gastrocnemius muscle after acute blunt contusion in Rabbits, observing the effects of electroacupuncture on Skeletal muscle morphological,the expression of Acetyl cholinesterase and Acetylcholine receptors. Researching the impact of electroacupuncture stimulation on the repair of skeletal muscle after contusion, to explore preliminarily the electroacupuncture treatment mechanism of skeletal muscle after acute blunt contusion.MethodForty-five adult New Zealand Rabbits (male or female,2.0-2.5kg) were made into heavy blunt injury model of gastrocnemius muscle of the right hind limb and were randomly divided into three groups:the model group, the electroacupuncture group and the positive control group,15animals in each group. The model group was not given any treatment; Rabbits in the electricacupuncture group were treated within24h after Contusion,which gone once every other day. Zusanli acupoint was in the uninjuried side, the depth of the positive electrode stabbing into Zusanli acupoint was15mm, the negative electrode were oblique insertion into subcutaneous near Zusanli acupoint5mm;Ashixue acupoint was10mm from the far and near end of the injuried area, the depth of positive electrode and oblique insertion stabbing into Ashixue acpoint was15mm. the electroacupuncture group was performed for15minutes every two days with0.4mA (2Hz);0.lmg/ml IGF-1,0.25ml per rabbit,was injected intramuscularly into the damaged gastrocnemius and gone every week in positive control group. Materials were picked up on the7th day,14th da yand28th day(n=5).HE staining was used to observe the general situation of muscle fibers, GENMED frozen section acetyl cholinesterase activity staining kits was used to observe the expression of AchE, fluorescent staining was used to observe the expression of AchR. Results1. After contusion of skeletal muscle, as the extension of time of injury (7d,14d and28d),the expression of AchE in each group is on the rise; The average diameter and area of the motor end plate include the expression of integrated optical density (IOD) of AchE at erery time point (7d,14d and28d) in positive control group (IGF-1) were higher than that of in electroacupuncture group (P<0.05), that of in the electroacupuncture group is higher than that of in the model group (P <0.05), which is statistically significant difference between groups.(as shown in figure6、7、8) 2. After contusion of skeletal muscle, as the extension of time of injury (7d,14d and28d), the expression of AChR in each group is on the rise; The average diameter and area and integrated optical density (IOD) of AChR at erery time point (7d,14d and28d) in the positive control group (IGF-1) is higher than that of in the electroacupuncture group (P<0.05), that of in the electroacupuncture group is higher than that of in the model group(P<0.05), which is statistically significant difference between groups.(as shown in figure10、11、12) Conclusion1. Electroacupuncture is beneficial to promote the restoration of the motor end plate of the damaged skeletal muscle, it may be associated with accelerating the expression of AChE and AchR.2. Electroacupuncture is contribute to promote the restoration the expression of AChE and AChR of the motor end plate of the damaged skeletal muscle,which may also be one of the mechanisms of repairing and regeneration of the damaged skeletal muscle, but the specific mechanism remains to be further research.
