Objective:To investigate the distribution of genotypes of TGFJ31T29C and evaluate the relationship between polymorphism of T29C in TGFJ31gene and susceptibility and radiosensitivity of non small cell lung cancer (NSCLC).Method:Polymerase chain reaction technology for122cases(60cases of non-small cell lung cancer and62cases of healthy people) of transforming growth factor β1T29C gene was amplified,PCR products were sequenced directly with ABI instrument.Analyse the genotyping situation of NSCLC group and the healthy control group and the correlation between genotypes and radiotherapy.60cases of lung cancer patients underwent radiation therapy, all patients did not given chemotherapy before radiation therapy.The total radiation dose was60Gy-70Gy,2Gy per day,5times per week.Specific dose based on individual circumstances.Analysis of the correlation between genotypes and radiotherapy. All tests were conducted at the P=0.05level of significance,using SPSS13.0software package and SHEsis software.Result:1.The test of genetic polymorphisms frequency distribution in control group and NSCLC group were in Hardy-Weinberg equilibrium law.T and C allele frequencies in NSCLC group and the control group were51.7%,58.1%and48.3%,41.9%respectively. TT,TC,CC genotype frequencies in NSCLC group were28.3%,46.7%and25.0%.TT,TC,CC genotype frequency in control group was33.9%,48.4%and17.7%,but the difference between the two groups is not significant(P>0.05).2.Effective rate of radiotherapy in NSCLC patients with heterozygous genotype(TC) was only28.6%,the mutant homozygotes genotype(CC) in NSCLC patients was50.0%,and a higher efficiency of64.7%in the wild homozygous genotype(TT) in patients with radiotherapy,and the difference was significant(χ2=6.125P<0.05).Conclusion:There is no significant relationship between the T29C SNP in TGFβ1and susceptibility in NSCLC.The TGFβ1T29C SNP is closely associated with radiosensitivity of NSCLC in the ethnic Han Chinese in Hunan province.
