Objective: This study was established by intragastric ice waterconstipation predominant irritable bowel syndrome rat model,The model weretreated by drugs of mazhijiangzhuo Decoction. To observe the generalcondition of rats,Fecal water content,The number of mast cells in rat ileocecal,inside the colon tissue. To investigate the MaZhiJiangzhuo decoctionprescription on treatment of constipation irritable bowel syndrome and itstreatment mechanism, thus syndrome provide a experimental basis andtheoretical basis for MaZhiJiangzhuo Decoction in treating constipation typeirritable bowel.Methods:(1)Baseline: Selection of clean grade healthy male40wistar rats. Weight140±20g. The independent feeding cage,collect manure. Adaptive feeding for7days, select normal rat test.(2)Manufacturing mode: Grouping: dividing animals into two groups i.e.A group and B group. Group A was the normal control group, a total of8, theexperimental stage and given normal feeding, drinking water, not gastriclavage. B group and model group with32rats,8:30every day gavage1times,giving the0-4℃2ml saline, molding period of14days,Observation in ratsafter intragastric administration of24h waste water content, and record.Measurement of body weight once every7days in rats. In the fecal grains andchange water for modeling success criteria.(3)The treatment period: After the success of modeling, the rats of Agroup normal feeding, drinking water, not gastric lavage. The B group wererandomly divided into4groups, with8rats in each group, Model controlgroup rats were given distilled water2ml/d, The western medicine group given cisapride3.6mg/kg/d, traditional Chinese medicine control group rats weregiven compound aloe capsule0.36g/kg/d, MazhiJiangzhuo Decoction group.Rats were treated with MaZhiJiangzhuo prescription6.3g/kg/d, interventiontreatment period of2weeks. Drug dosage according to body surface areaconversion, To observe the changes of gastric perfusion,24h rats fecal watercontent and record, measurement of body weight once every7days in rats, thebody weight changes of the rats were observed.(4) Evaluation period: after the last administration, fasted24h. Rats werekilled specimens were taken. Quickly open the whole gastrointestinal tissueremoved after death, to measure the distance to the pylorus ileocecal. Thenquickly take the rats ileocecal, colon (anus5cm) each0.5cm, rinseimmediately with saline, l0%neutral formalin fixative fixed, paraffin-embedded sections. In the light microscope intestinal tissue pathology, takeileocecal and colon of rats paraffin sections, each taking three patients per site,MC toluidine blue staining using improved staining was observed under anoptical microscope, performed MC count.Results:(1)The general condition of rats:Rats with no light dry hair color, mentalirritability, no limb deformity, decrease of eating excrement form tends toelliptic globose, brown or gray white, hard, long uneven, long0.4-0.6cm,suggesting the model is successful. Intervention treatment of rat hair color toshine, nervous feeling better, no limb deformity, food intake increased, stoolshape tends to normal, brown, long approximately0.7cm. Treatment ofprompt Si Shah Billy and Ma trifoliate compound aloe capsule,MazhiJiangzhuo Decoction on rat model of IBS-C was effective.(2) Pathological morphology of intestinal tissue: the structure of intestinalmucosal epithelial tissue integrity, cells arranged in neat, no organic disease,more in line with the physiological and pathological changes of the humanbody.(3)General observation of MC: after dyeing, optical microscope visibleMC distributed in the submucosa and lamina propria; cytoplasm by toluidine blue to purple red, the nucleus is blue, see section MC showed degranulationstatus, cell morphological changes.(4) MC count: model control group, western medicine group, Chinesemedicine group and the Mazhijiangzhuo Decoction group. Rats of ileocecalMC counts were compared to the normal control group (P<0.05); controlgroup of Western medicine, Chinese medicine group and the MazhijiangzhuoDecoction group. The rat colon were compared to the model of MC countcontrol group decreased (P<0.05); control group of Western medicine,Chinese medicine group and the Mazhijiangzhuo drop between the turbid ratsileocecal MC count were compared and no significant difference (P>0.05).The model control group, western medicine group, Chinese medicine groupand the Mazhijiangzhuo Decoction group. The rat colon MC counts werecompared to the normal control group (P<0.05); control group of Westernmedicine, Chinese medicine group and the Mazhijiangzhuo Decoction group.The rat colon MC counts were compared with model control group decreased(P<0.05); control group of Western medicine, Chinese medicine group andthe Mazhijiangzhuo drop between the turbid group rat colon MC count werecompared and no significant difference (P>0.05).Conclusions: After the modeling phase, group B rat droppings observedmorphological smaller than before and its water content is reduced, intestinaltissue cells were observed under a conventional pathology neat, tissue integrity,no organic change, suggesting that successful model. After treatment, themorphology and fecal water content tend to be normal, but with a normalcontrol group of rats contrast there are still some differences, promptintervention shorter treatment period, has not yet reached a cure. Back ratmodel group ileocecal and colonic tissue MC count increased compared tonormal control group, suggesting that there is a close relationship between theincidence of MC with IBS, but after each treatment group therapy interventionto reduce the number of MC compared with model group, suggesting hemporange drop cloud parties each control drugs can reduce the number of MCileocecal and colonic tissue, suggesting that the treatment may be associated with reducing the number of intestinal tissue to improve gut feeling MCabnormalities.
