Zanthoxylum armatum DC. is one of the genus of Zanthoxylum Linn. It contains many kinds of chemical constituents, such as lignans, triterpenes, amides, coumarins, alkaloids and flavonoids. It is a good medicine for stomachache, toothache and cold and it can be used to prevent snake bites. In this paper, the chemical constituents of ethyl acetate extract of Zanthoxylum armatum were isolated and identified, response surface methodology was used to optimize the ultrasonic-assisted aqueous two-phase extraction of lignans from Zanthoxylum armatum. It was also studied that the a-glucosidase inhibitory activity of Zanthoxylum armatum in vitro experiment. The main results were as follows:1.Five compounds were isolated from Zanthoxylum armatum. They were (-)-Fargesin, Sesamin, L-asarinin,3,4-dimethoxy-5-methyl-benzoic acid, butyl3,4,5-trimethoxybenzoate.(-)-Fargesin, Sesamin, L-asarinin are lignans.2.An aqueous two phase system (ATPS) was employed as the solvent to extract lignans from Zanthoxylum armatum. The result was optimized and projected by response surface methodology (RSM) and artificial neural network (ANN). The three standarded lignans (-)-Fargesin(Far), Sesamin(Ses) and L-asarinin(Asa) were used as marker compounds. The recoveries(Ro) of three markers were0.9910,0.9832and0.9870using ATPS with20%n-propanol and24%(NH4)2SO4. The RSM results showed liquid/solid ratio15:1, temperature40℃and time55min were the optimal ultrasonic-assisted extraction conditions, under these conditions,the purities of three markers reached2.222%,1.066%and1.583%with an increase of44.38%,26.34%and24.24%. The R (0.9855) and MSE (0.0018) values of model suggested good fitness and generalization of ANN. The results showed Ultrasonic-assisted ATPS method are suitable for extraction of lignans from Zanthoxylum armatum.3.The test of a-glucosidase inhibitory activity in vitro showed that the total saponins, alkaloids and polysaccharide had certain inhibition effect on a-glucosidase activity and the IC50values of a-glucosidase inhibitory effect were0.926,1.485,1.592, respectively.
