Zanthoxylum armatum DC. is one of the genus of Zanthoxylum Linn. Z. armatum contains kinds of chemical constituents, such as lignans, alkaloids, amides, flavonoids, triterpenes and steroids, coumarins, volatile oils, fatty acids and other chemical components. Z. armatum has the pharmacological activities of hepatoprotective, anti-inflammatory analgesic, anthelmintic, antispasmodic, and inhibition of platelet aggregation. In this paper, the optimization of ultrasonic assisted aqueous two-phase system extraction technology of total alkaloids from Z. armatum was studied. The ADP-induced platelet aggregation by the method of Born’s and anti-coagulation tests were researched. In addition, the α-amylase inhibitory effects of Z. armatum extracts was also studied. Furthermore, the isolation and identification of effective chemical constituents from Chloroform portion of the Z. armatum were studied. The main results were as follows:1. The ultrasonic extraction technology coupling with propyl-alcohol ammonium sulfate aqueous two-phase system for extracting total alkaloid from Z. armatum was analysed by the response surface methodology. The optimal extraction rate of total alkaloid from Z. armatum were extraction time40min, mass ratio of alcohol to water0.5:1, salt dosage7.79g. Under these optimized conditions, the extraction rate of total alkaloid was up to1.704%, which was higher than the traditional method. Analysis of variance showed that the salt dosage, mass ratio of alcohol to water had an extremely significant impact on the extraction of the total alkaloid, and the impact of extraction time on the extraction rate of total alkaloids is significant.2. The result of the ADP-induced platelet aggregation by the method of Born’s indicate that the four components which were extracted from Z. armatum could significantly inhibit platelet aggregation except the polysaccharide. Besides, the effect on blood coagulation was test by the indicators of PT, TT, APTT, and the results showed that a portion of components had significant anticoagulant activity.3. The results of the α-amylase inhibitory effects of Z. armatum extracts indicated that the inhibition on α-amylase activity of polysaccharides was not obvious, but the saponins had significant inhibition activity, which was higher than acarbose.4. Seven chemical constituents were isolated from Z. armatum:compound Z1(syringic acid), compound Z2(L-asarinin), compound Z3(β-sitosterol), compound Z4(horsfieldin), compound Z5(eudesmin), compound Z6(L-sesamin), compound Z7(fargesin).
