Aims:To study the protective effect of limb remote ischemic preconditioning (RIPC) against renal ischemia reperfusion injury and discuss mechanism against renal ischemia reperfusion injury in mice.Methods:Fifteen male mice, weighing20-25g, were randomly divided into three groups:sham operation group (Sham), ischemia reperfusion group (I/R) and limb remote ischemic preconditioning group (RIPC),five mice in each group. Opened the abdominal cavity, separating the left side of the renal pedical but no occlusion was perfomed, then animals were observed for35min in Sham group; in I/R group, with the use of a nontraumatic vascular clip,35min of left renal artery occlusion was induced followed by24h of reperfusion, and underwent right nephrectomy. Limb remote ischemic preconditioning was induced by a rubber band, the animals underwent four episodes5min ischemia and5min intervals, then left renal artery was accluded for35min followed24h reperfusion, and underwent right nephrectomy in RIPC group. Blood samples from orbital were taken for serum creatinine (Scr) and blood urea nitrogen (BUN) after24h reperfusion. The serum TNF-a, IL-6, IL-10and concentrations were measured by luminex. Then the animals were killed and the left kidneys were removed for histological examination by hematoxylin and eosin (HE).Results:24h after reperfusion, Compared with Sham group, serum creatinine and blood urea nitrogen, and the score of tubular damage were significantly increased in I/R group (p<0.05). The serum concentration of TNF-α and IL-6were significantly increased while serum concentration of IL-10had no significant difference between these two groups. Compared with I/R group, serum creatinine and blood urea nitrogen, and the score of tubular damage were significantly reduced in RIPC group. The serum concentration of TNF-α and IL-6were significantly reduced while serum concentration of IL-10was significantly increased in RIPC group (p<0.05).Conclusion:Limb remote ischemic preconditioning has protective effect against renal ischemia reperfusion injury in mice. Limb remote ischemic preconditioning may inhibit the expression of TNF-α, IL-6, and increase the expression of IL-10, so as to play a role in renal protection by balancing the anti-inflammatory and inflammatory reaction of immune function.
