Objective:We detected the expressing of the vascular endothelial growth factor (VEGF) and matrix metalloproteinases-2(MMP-2) in osteosarcoma tissues, and analyzed the relationship with clinicopathological parameters and between them, which wil lay the foundation for the study of the pathogenesis of osteosarcoma.Method:1, Case selection:All cases of osteosarcoma sample were from patients by surgical treatment in the affiliated Xiang ya Hospital of Central South University frome2008to2011,15normal cases were selected as control rroup;2, HE staining for further validate osteosarcoma;3, Detecting expression of MMP-2and VEGF levels in osteosarcoma and normal tissues by S-P immunohistochemical staining.Results:1, MMP-2was highly expressed in osteosarcoma compared with normal bone tissue with low expression, in osteosarcoma tissue, the positive expression rate of MMP-2was65.71%.In normal bone tissue, the positive expression was20%.There are significant difference in the two groups; MMP-2expression was no significant difference with patient’s age, sex in osteosarcoma; The positive rate of MMP-2expression of osteoblast type was60.00%, chondrocytes type was63.64%,and fibroblasts type was66.67%.There are no significant difference among the three groups; The positive rate of MMP-2expression in high and middle grade was42.00%, and low differentiated grade was78.26%.There was a statistically significant difference; MMP-2expression rate was79.16%in soft tissue invasion samples,and36.36%without soft tissue invasion. There are significant difference;The positive expression rate of MMP-2in postoperative pulmonary metastasis of osteosarcoma was100.00%, and47.83%without lung metastasis.The difference was significant.2, VEGF presented high expression in osteosarcoma and low expression in normal bone tissue. The positive expression rate was74.29%in osteosarcoma, and20%in normal bone tissue,which difference was significant. VEGF expression was no significant difference with patient’s age, sex in osteosarcoma; The positive of rate VEGF was73.33%in osteoblast type,81.82%in chondrocytes type,66.67%in fibroblast type,which showed no statistically significant difference among the three groups; VEGF expression in high and middle grade was50.00%, and the low differentiation was86.96%, which showed a statistically significant difference; VEGF expression rate was87.50%in soft tissue invasion,and45.45%without soft tissue invasion,which showed statistically significant difference; The positive expression rate of VEGF in pulmonary metastasis of postoperative in osteosarcoma was100.00%,and47.83%without lung metastasis.The difference was significant.3, MMP-2and VEGF showed high expression in osteosarcoma,and a Significant positive correlation between the two factors.Conclusion:1, MMP-2and VEGF showed high expression in osteosarcoma. The positive expression rate in osteosarcoma were significantly higher than in normal bone tissue.MMP-2and VEGF may play an important role in the incidence and development of osteosarcoma.2, MMP-2and VEGF expression were significantly higher in osteosarcoma of soft tissue invasion and postoperative pulmonary metastasis. We infer that MMP-2and VEGF play an important function in the local infiltration and distant metastasis.3, MMP-2and VEGF expression in osteosarcoma presented obvious relevance. MMP-2and VEGF may become the potential gene treatment target of osteosarcoma.
