Background: OGT originally found in the liver, and its expression levelrelates to cellular functions.OGT is the only glycosyl transferase to theO-GlcNAc glycosylation. O-GlcNAc glycosylation is defined as the covalentattachment of a single N-acetylglucosamine molecule to serine or threonineresidues, is a posttranslational modification of intracellular proteins. AbnormalO-GlcNAc glycosylation is closely related to wide varieties of tumors,consider OGT expression level also changes in the tumors.Objective: To screen of the expression and clinical significance of OGT inhepatocellular carcinoma.Methods: Western blot was used to detect the expression of OGT in20samples of HCC and matched non-tumor tissues, meanwhile to detect theexpression of OGT in8kinds of hepatoma cell line and normal liver celllysate.OGT expression was detected by direct ELISA in serum specimens(healthy volunteers and patients with LC, HCC) in20cases.Results: There was expression of OGT in both HCC and matchednon-tumor tissues in western blot.And in HCC tissues,the total levels of OGTHCC proteins were significantly higher than that in the non-tumor tissues (P<0.05).And there was expression of OGT in both hepatoma cell line and normalliver cell lysate in western blot. And in hepatoma cell line, the total levels ofOGT HCC proteins were significantly higher than that in the normal liver celllysate(P<0.05).The result of direct ELISA showed that OGT was highlyexpressed both in serum specimens of LC and HCC patients than that inhealthy volunteers(P<0.05).Conclusion: OGT was overexpressed in HCC and OGT can be a useful diagnostic value for HCC.
