Objective: To investigate a novel way from salinomycin, an inhibitor ofwnt signal transduction way to suppress triple negative breast cancersubpopulation enriching stem cells.Methods: Triple negative breast cancer HCC38cell lines with positiveALDH1expression was incubated in vitro. The cells treated by NormalSaline,Salinomycin and Purple calcitriol were divided into vehiclegroup、observed group and positive control groups.1.MTT was used to assay the growth of HCC38cell line.2.flow cytometry was used to detect cell apoptosis.3.Microspheres (mammospheres, MSs)formation was applied tforidentifying the exist of stem cells.4.Transwell invasion assay proved tumor cell migration.5.Westen blot test for detection of β-catenin protein levels.Results: HCC38CELLS can form microsphere.Compared withvehicle group, Salinomycin able destruction sphere, and compare withpurple calcitriol salinomycin more obvious,can inhibit cell growth,induce apoptosis and reduce cell invasiveness. Reduced proteinexpression of wnt down-stream in HCC38. Conclusion: The inhibitor of wnt signal transduction pathways may be ofpotential treatment value against triple negative breast cancer enrichingstem cells through inhibit the down stream protein of wnt.It might alsobe prospective inhibitor of breast cancer stem cells.
