Objective: To study the expression and significance of NDRG1,COX-2and VEGFin cervical cancer tissue and to evaluate the effect and mechanism of NDRG1onproliferation and apoptosis of cervical cancer cells and to provide evidence for diagrosisand prognosis of cervical spuamous cancerMethods: NDRG1, COX-2and VEGF expressions were detected byimmunohistochemistry (IHC) in15cases of normal cervical tissue and in80cases ofcervical spuamous cancer tissue. The correlation among NDRG1, COX-2and VEGFexpressions in cervical cancer tissue was investigated by Spearman rank correlation analysis.The relationship between NDRG1expression and clinicopathological characteristics wasinvestigated Kaplan-Meier survival curve analysis. pcDNA3.0-NDRG1and pcDNA3.0were transfected into SiHa cells through lipofectamine and positive clones were screenedby G418. Reverse Transcription Polymerase Chain Reaction (RT-PCR) and western blotwere performed to identify mRNA and protein expressions of NDRG1in SiHa cellsrespectively. The expressions of COX-2and VEGF in positive clones were detected byRT-PCR and western blot. The cell proliferation and apoptosis were detected by MTTassay and flow cytomertry.Results:(1) In tissue samples from80cervical cancer cases, positiveimmunoreactivity expression of NDRG1,COX-2and VEGF were52.2%,91.2%and77.5%respectively. Positive rate of NDRG1was significantly lower than those in normal cervicaltissue (P<0.01). However, positive rate of COX-2and VEGF was higher than those innormal cervical tissue(P<0.01). There was no significant difference between positive rateof NDRG1,COX-2and VEGF expressions with clinicopathologic data of cervical cancerpatients including age and tumor size(P>0.05). Positive rate of NDRG1,COX-2andVEGF expressions was significant correlated with tumor histopathological grade,clinicalFIGO stages and lymph node metastasis(P<0.01). There was statistically positivecorrelation among NDRG1,COX-2and VEGF expressions in cervical cancer tissue (P<0.01).(2)There was statistically correlation between positive rate of NDRG1,COX-2and VEGF expression with survival time by Kaplan-Meier analysis.Conclusion:The expression of NDRG1in cervical cancer tissue was down-regulatedand the expressions of COX-2and VEGF in cervical cancer tissue were up-regulated.NDRG1may play an important role in the development and metastasis of cervical cancer.The joint detection of NDRG1,COX-2,VEGF may provide some guiding significance tojudge the prognosis of cervical cancer..(2)NDRG1transfection could effectively promotethe apoptosis and inhibit the proliferation of SiHa cells by downregulating COX-2,VEGFexpression in SiHa cells.
