The pelvis and acroteric soft tissue are one of predilection site of connective tissue tumor. Because earlier period symptoms are not obviously, malignant tumor in pelvis have in middle or advanced stage when it can be found, the tumor is large and adhere with acroteric soft tissue. The main therapy method of malignant tumor in pelvis is operation. Compare with orthodox amputation, protect-limbs-operation of pelvis is important.To cut tumor thoroughly is the condition of protect-limbs—operation of pelvis and curing malignant tumor. If femoral nerve or sciatic nerve is be offended, the protect-limbs-operation of pelvis will be quit.At present, a positive nerve’s method of Allograft is transplantation after Freezing. In this experiment,homogeneity variant nerves depressed Immunogenictiy by liquid Nitrogen. On this basis, research the physiologic function with different length neurologic defect after repairation, and have a instruction of enlarging the applicability of protect-limbs-operation and reducing the recurrence of turmor.Objective:In this experiment, we set up the SD rat model through injuring sciatic nerve, under microsurgery, we cut different length sciatic nerve, transplant with the same length Freezing homogeneity variant nerves by liquid Nitrogen. We observed the function of transplantation after operation by footprint and morphology, and provided experimental data of enlarging the applicability of protect-limbs-operation and reducing the recurrence of turmor.Methods:60rats SD rats were randomly divided into12groups (n=5):variant nerve transplantation group with the length of10mm(A1),15mm(A2),20mm(A3),25mm(A4); Freezing nerve transplantation group with the length of10mm(B1),15mm(B2),20mm(B3),25mm(B4); Hepatitis nerve transplantation group with the length of10mm(C1),15mm(C2),20mm(C3),25mm(C4); During each group, we were randomly divided into4groups by the length of absent sciatic nerve(10mm,15mm,20mm,25mm). And under10times operating microscope, rats were transplanted with the homologous group and variant nerve,Freezing nerve and hepatitis nerve. We determined the different groups rats’sciatic nerve function index(SFI: Significance of SFI:-100means injury completely,0means nulli-injury.) and wet weight of gastrocnemius(12weeks after operation, we took rat’s gastrocnemius, marked the weight of right divided the weight of left), observed the different groups rats’neurofilament protein(NFP) by immunohistochemistry(We observed transplantation’s distal end by SP immunohistochemical stain, found that the brown corpuscular quantity in sciatic nerve’s auxiliary fibers could express neural regeneration’s quantity).Results:1.SFI:B3group’SFI (-80.49±0.3) had statistical significance with C3group’ SFI (-73.27±2.41)(p<0.05)2.Wet weight of gastrocnemius(%):A3group (25.71±1.90) had statistical significance with B2group (41.79±6.46) and C2group (56.78±1.85)(p<0.05);B4group (24.62±7.65) had statistical significance with C4group (30.09±1.52)(p<0.05)3.NFP:B4group(55.33±9.02) had statistical significance with C4group(682.67±147.80)(p<0.05);A3group(85.33±36.95)had statistical significance with B3group(395.00±48.57) and C4group (682.67±147.80).(p<0.05)4.Results of Transmissiong Electron MicroscopeA3group:there are rare Schwann wells with thin endochylema, uncomplete caryotheca and amyelination.Bindweb’s hyperplasy are obviously. B3group:there are less Schwann wells with tenuity endochylema, fairly complete caryotheca and less bioblast and endocytoplasmic. C3group:there are much Schwann wells with satiation endochylema, complete caryotheca and plenty of bioblast and endocytoplasmic and complete medullary sheath.Conclusion:Through this experiment,rat’s sciatic nerve was cut inside20mm, nerve regeneration eaqual autoimmune neural transplantation by freezing allosome transplantation. We believe that this idea can provid a new way of operation of malignant tumor of pelvis.
