Objective: To observe the different donor liver cold preservation time limit effect on bileduct epithelial cells apoptosis; Detection of apoptosis related proteins the Bcl-2and Fas indifferent cold preservation time limit for the change of hepatic duct epithelial cells express;To explore the Bcl-2and the meaning of Fas in bile duct epithelial cells apoptosis; Exploringunder the cold storage time limit for the safety of the liver, in order to reduce the livertransplantation biliary complications, improve the success rate of liver transplantation.Methods:36healthy male SD rats, random points6groups, each group of6, not coldperfusion1group is set to the control group, according to the donor liver perfusion coldpreservation time points after5groups respectively for1h,3h,6h,12h and24h group. UWfluid infusion after abdominal aorta and common bile duct, living in liver tissue preservationin the UW solution of4degrees Celsius, according to the grouping save different timelines.Microscope and transmission electron microscope bile duct epithelial cells and mitochondria,endoplasmic reticulum and golgi apparatus in the cell organelles of the change;Immunohistochemical method to detect bile duct epithelial cells apoptosis related proteins theBcl-2, and the expression of Fas.Results:1.The microscope results: control group,1h,3h group were no obvious changesof bile duct epithelial cells of the cubic,6h group, bile duct epithelial cells of the cubicappeared mild swelling,12h bile duct epithelial cells of the cubic group a swelling, existcytoplasm vacuoles, cell nucleus appeared mild increase,24h group of bile duct epithelialcells of the cubic swelling, cytoplasm vacuoles, cell nucleus appeared swollen, the portal areaappear a small amount of inflammatory cell infiltration;2.Electron microscopy (sem)observation, the control group,1h,3h group, bile duct epithelial cells of the cubic surfacevillus is clearly visible, neat rows, there are a large number of endoplasmic reticulum andmitochondria in cytoplasm, there is a small amount of golgi apparatus and endoplasmicreticulum, mitochondria, golgi morphology neat, no obvious changes,6h bile duct epithelialcells of the cubic group a slight disturbance, the surface of the villi is rich, relatively less thancontrol, cytoplasmic mitochondria, endoplasmic reticulum and golgi apparatus in form neat,no obvious change,12h bile duct epithelial cells of the cubic group villi, arrange a milddisorder, the cytoplasm of mild swelling of mitochondria, endoplasmic reticulum expansion,and golgi apparatus,24h group of bile duct epithelial cells of the cubic surface villus decreased, disordered arrangement, part of mitochondrial crest breaking, some mitochondriavacuoles change, endoplasmic reticulum swelling is severe, part of the endoplasmic reticulumin vacuoles, a small amount of endoplasmic reticulum appear rupture, golgi apparatus, mostof the expansion of a flattened sacs, accompanied by flattened sacs, big and small dip splitdissolved;3.Immunohistochemical results: the Bcl-2in the control group, group1h,3h,6hgroup in bile duct epithelial cells of the cubic expression with low, high expression of12hgroup,24h group of lower expression; Fas in the control group, group1h,3h,6h group in bileduct epithelial cells of the cubic expression is low, high expression of12h group,24h groupof lower expression.Conclusion:1.As the cold storage time prolonging, bile duct epithelial cells less ofischemia and hypoxia tolerance of liver cells, prompt changes in bile duct epithelial cells inliver transplantation sooner in liver cells.2.Shortening the time of donor liver cold storage,can effectively reduce the incidence of biliary complications after liver transplantation,12hwithin the safety of the time limit may be liver transplantation.3.Apoptosis related proteins inliver transplantation for hepatic cold preservation injury of bile duct cells may play animportant role.
