Experimental Study On Protection Of Gastric Mucosa In Irritability Gastric Ulcer Rats By Zuojin Pills Of Hepatogastric Tongzhi Method

Objective By observing the rat gastric body morphology, gastric juice PH value and calculation were gastric ulcer index (UI), judge the protection of Zuojin pills which reflect the Hepatogastric Tongzhi method on irritability gastric ulcer rat gastric mucosa. And by using enzyme-linked immunoassay detection content changes of ACTH、 CORT、SOD、MDA、 PGE2in irritability gastric ulcer rats serum, comparing rat gastric mucosal tissue HE staining microscopy results, to preliminary discussion the mechanism of the protection of Zuojin pills which reflect the Hepatogastric Tongzhi method on irritability gastric ulcer rat gastric mucosa.Methods Sixty healthy male Sprague-Dawley rats were randomly and equally divided into blank control group, model control group, positive control group, and low-,middle-,high-dose Zuojin pill groups. The rats received respective treatments by prophylactic administration for5d:the blank control group and model control group received normal saline by gavage; the positive control group received water suspension of omeprazole by gavage; the low-, middle-, and high-dose Zuojin pill groups received low-, middle-, and high-dose water decoction of Zuojin pills by gavage. After the last administration, followed by24h of fasting (without water deprivation), a rat model of gastric ulcer was established by water immersion-restraint stress in all groups except blank control group. After the model was established, gastric morphology was observed; the PH value of gastric juice was measured; gastric ulcer index (UI) was calculated; serum levels of ACTH、CORT、MOD、SDA、PGE2were determined by enzyme-linked immunosorbent assay, and the gastric mucosal tissue HE staining microscopy results was compared.Results1. Compared with blank control group, model control group rats gastric juice PH value decrease, a significant rise in the UI(P<0.01); Compared with model control group, middle-and high-dose Zuojin pill groups rats gastric juice PH value rise obviously, a significant decrease in the UI(P<0.01); Compared with positive control group, high-dose Zuojin pill group rats gastric juice PH value and UI have no obvious difference(P>0.05).2. Gastric mucosal tissue HE staining microscopy results:Blank control group in stomach tissue structure is complete, no damage; Compared with blank control group, there was an obvious impaired gastric mucosal epithelium、fall off, vascular congestion obviously, and even a large number of bleeding in model control group; Compared with model control group, there was defect and effusion on gastric mucosa in positive control group and middle-, high-dose Zuojin pill groups, but the lesion was significantly lighter.3. Compared with blank control group, there was a significant rise in the levels of ACTH and CORT in model control group rats serum(P<0.01); Compared with model control group, there was a significant decrease in the levels of ACTH and CORT in positive control group and high-dose Zuojin pill group rats serum(P<0.01), and also in middle-dose Zuojin pill group(P<0.01or P<0.05); Compared with positive control group, middle-dose Zuojin pill group and high-dose Zuojin pill group rats serum ACTH and CORT levels have no obvious difference(P>0.05).4. Compared with blank control group, there was a significant rise in the level of MDA and a significant decrease in the level of SOD in model control group rats serum(P<0.01); Compared with model control group, there was a significant decrease in the level of MDA and a significant rise in the level of SOD in positive control group and high-dose Zuojin pill group rats serum(P<0.01), and also in middle-and low-dose Zuojin pill group(P<0.01or P0.05); Compared with positive control group, middle-dose Zuojin pill group and high-dose Zuojin pill group rats serum MDA and SOD levels have no obvious difference(P>0.05). 5. Compared with blank control group, there was a significant decrease in the level of PGE2in model control group rats serum(P<0.0l); Compared with model control group, there was a significant rise in the level of PGE2in positive control group and high-, middle-dose Zuojin pill group rats serum(P<0.01); Compared with positive control group, middle-dose Zuojin pill group and high-dose Zuojin pill group rats serum PGE2levels have no obvious difference(P>0.05).Conclusion1. Zuojin pills which reflect the Hepatogastric Tongzhi method can protect the gastric mucosa of irritability gastric ulcer rats.2. Zuojin pills through increased gastric juice PH value and improve the PGE2levels in serum, to reduce gastric mucosa against factors and improve the ability of gastric mucosa defense, So to protect the rat gastric mucosa.3. Zuojin pills by improve the level of SOD and decrease the level of MDA in serum of rats, to improve the ability of the body resistance to oxidation and scavenging free radicals, in order to play a protective role to gastric mucosa of irritability gastric ulcer rats.4. Zuojin pills by reduce the level of CORT and ACTH, to suppress the secretion of gastric acid and promote the renewal of the gastric mucosa, in order to play a protective role to gastric mucosa of irritability gastric ulcer rats.
