The Influence Of Electro-acupuncture Mingmen On Ovariectomized Rat Hypothaiamus BMP-2and Its Signal Transduction Protein Smad1/5Expression | Posted on:2015-01-23 | Degree:Master | Type:Thesis | Country:China | Candidate:W Qin | Full Text:PDF | GTID:2254330428970825 | Subject:Acupuncture and Massage | Abstract/Summary: | PDF Full Text Request | Objective:To study the bone morphogenetic proteins BMP-2and its signal transduction protein Smadl/5expression in the hypothalamus of ovariectomized rats model with electro-acupuncture(EA) at Mingmen Point (GV4). Discussion EA Vital Points treatment of postmenopausal osteoporosis mechanism. Methods:sixty3.5-month-old female SD rats were randomly divided into sham operation group, control group, EA Mingmen group(Mingmen group), EA non-acupoint group(non-acupoint group),(n=15). In addition to the sham group, the rats were ovariectomized, after all animals reared under the same conditions after90days of intervention, Mingmen group and non-acupoint group EA1times daily, each lasting20min (10times as a course) were treated three courses; control group and sham group given the same daily crawl of the above-mentioned groups, fixed.3, after the end of treatment, take the rat hypothalamus, Using light microscopy to femur bone morphology observed;using immunohistochemical methods, measuring hypothalamic BMP-2and its signal transduction protein Smadl/5expression.Results:1. Femoral bone morphology analysis:Trabecular color image comparison:Sham group compared with trabecular arrangement rules intensive, more uniform distribution; Model group compared with sparse fine trabecular spacing widens the gap some areas appear larger; EA group Vital Points dense trabecular thickening, narrowing the gap; EA non-acupoint group compared with sparse fine trabecular spacing widens the gap larger part of the region appears.2. Hypothalamic expression analysis of BMP-2and its signal transduction project Smadl/5.Using an image analysis software hypothalamus BMP-2and its signal transduction protein Smadl/5positive cells in the average gray value. BMP-2expression levels①Mingmen group was significantly higher than the model group and non-acupoint group (P <0.05); levels close to the sham group (P>0.05).②By non-expression of BMP-2was significantly lower than non-acupoint group and the sham group Vital (P<0.05); levels close to the model group (P>0.05)Conclusions:1.Electro-acupuncture Mingmen Point can significantly increase rat hypothalamus BMP-2expression and signal transduction protein Smadl/5.2.Electro-acupuncture Mingmen Point can prevent postmenopausal osteoporosis. | Keywords/Search Tags: | ovariectomized, Mingmen(GV4), BMP-2, Smad1/5, thehypothalamus | PDF Full Text Request | Related items |
