Valeriana amurensis is the drying root and rhizome of Valeriana amurensis which belongs to Valerianaceae and Valeriana. Based on the previous research of our group, the petroleum ether extract of Valeriana amurensis has significant effect on the improvement of sleep, of which8compounds including kissoone B show remarkable active effect selected by Drosophila melanogaster method. Based on this, more research will be carried out to study the effect of kissoone B on the sleep of Drosophila melanogaster further and its sleep mechanism on the molecular level.Kissoone B compound of5different kinds of concentration (0.25%,0.5%,1%,2%,4%) and7different medication period (1d,2d,3d,4d,5d,6d,7d) is studied to obtain corresponding effect on the sleep time of7-day-old Drosophila melanogasters by DAMS monitoring system; the Drosophila melanogaster lonely model is set up to investigate the effect of the monomer compound kissoone B on its sleep time and prove its pharmacological activity of improving sleep; Taking four expression variables of Drosophila melanogaster brain per, tim, clk, slo as indexes, the change of gene expression variables corresponding to the sleep of7-day-old Drosophila melanogasters is studied before and after the usage of kissoone B.The experimental results show:1. The effect of kissoone B of different concentration on the sleep of7-day-old Drosophila melanogasters:compared with the blank group, significant differences concerning daytime sleep time, night sleep time and total sleep time arise except the group of0.25%(p<0.05)2. The effect of kissoone B of different medication periods(1-7days) on the sleep of7-day-old Drosophila melanogasters:the sleep time of male Drosophila melanogasters increase after2-day medication (p<0.05), and increases significantly after5-day medication (p<0.01). The total sleep time of male Drosophila melanogasters also increases remarkably after5-day medication compared with2-day group (p<0.05). The total sleep time of female Drosophila melanogasters increases remarkably after3-day medication compared with the control group (p<0.01)3. Experimental results concerning the effect of the amount and time of kissoone B usage on the sleep of Drosophila melanogasters show that the best concentration for male and female Drosophila melanogasters is1%and0.5%respectively and the best medication periods are3days and5days respectively.4. Kissoone B can extend the sleep time of lonely-mode Drosophila melanogasters significantly.5. The gene expression variables per、tim、clk and slo are all increased by adding valerian petroleum ether extract; The gene expression variables per、clk and slo are all increased by adding kissoone B while tim of male ones is also increased.Therefore, the effect of the petroleum ether extract of Valeriana amurensis and kissoone B on the sleep time of Drosophila melanogasters is the result due to the process involving multiple genes and multi-gene joint regulation.
