It frepuently used traditional Tibetan medicine plant from an area of Tibet where is called La Gang, a different name is Ge Tu Man Ba and Xin Mu Tou Man Ba, etc. Geranium pretense L. is non-genuine Medicinal material, and the "ChP" and "Tibetan ministerial" unrecord. It has been recorded in the "Chinese herbal medicine· Tibetan Volume" and "Tibetan". It’s mainly growns in shrubs and bushes, the alpine meadow, under the tree or the riverside of the lawn, at the elevations between2600m to4000m. It’s mainly distributed in northwestern Sichuan, southeastern Tibet and northeastern Qinghai. It shows the effects of the lee lung chills, inflammation diarrhea, drainage and detoxification effects. That’s mainly used enteritis and diarrhea, typhoid, sound off, indigestion, etc.In this paper, it’s conducted a Geranium pretense L. textual research, and the macroscopic macroscopic morphology of plants which is study to the structure of stems, petioles and leaf, and were Pharmacognosy identification system from which the microstrucrure of the powder characteristics the plants:7to9leaves, rhombic or small rhombic, trichomes on calyx. The anatomical features, it thick that stems of cells belts were11to12, and the petioles of cells belts were5to6. This medicinal properties, anatomical features and powder characteristics is the difference with the other plants.This paper, choice the both shikimic acid and gallic acid as standard that nine batches of samples were TLC qualitative identification with dichloromethane-ethyl acetate-methanol-methanoic acid (3:4.5:0.5:1) and cyclohexane-ethyl acetate-methanoic acid (8:6:1) as the agent, and humidity of40%to80%, then, the both and other components well separated, Rf were0.51and0.39. The results show that the methods is simple, rapid, accurate sensitivity, the results of shikimic acid and gallic acid linear regression equation were y=42250x+17303, r=0.9995; y=3.5946×106x-21568, r=0.9999; the ranges were124.5~1992.0μg,0.064~0.448μg; the average recovery was99.74%and102.96%; RSD 1.68%and0.58%. And then, determination of the nine batches from G. pretense L. herbs that were shikimate and gallic acid of content, the results show, the content of shikimic acid was0.16%to0.23%, the content of gallic acid was0.18%to0.38%.In the work, this paper drafts a draft the medicinal of quality standards. Preliminary, medicinal of content calculated on dry goods, shikimic acid content of not less than0.15%, gallic acid content of not less than0.20%; And medicinal of moisture not more than12.0%, the total ash content not more than12.0%, acid insoluble ash not more than3.0%.In this paper, the maximum absorption wavelength of UV was established to determine the total tannin in Geranium orientali-tibeticum R. Knuth, gallic acid was used as reference, and phosphotungstomolybdate acid-casein was used as colour-developing agent. The results show that this method is rapid, simple, accurate and sensitive. Gallic acid linear regression equation was y=112.28x+0.0081, r=0.9998. The average recovery was99.29%, RSD1.22%. The total tannin content determination by this method was8.70mg· g-1. This methods not only as the medicinal plants tannin content determination process of evaluation, but also learn from the experience and provide a theoretical reference for other medicinal plants.The traditional ethanol extraction from Geranium orientali-tibeticum R. Knuth was optimized by using orthogonal test method. The optimum extracting conditions were eight times quantity of70%ethanol, three times of extraction,1h a time. The rate of Tannin extraction was84.95%. The contant of Tannin was up to10.20%. The contant of Tannin was7.23%.The extraction of total tannin from Geranium orientali-tibeticum R. Knuth material by supercritical influence CO2extraction was studied. Taking the conent of total tannin as the evaluating condition, the supercritical influence to extraction major factors such as the extraction pressure, the extraction temperature, the extraction time and influence flow rate of CO2were investigated. The optimum condition for supercritical influence CO2were obtained by orthogonal experiment design, the optimum condition was followed:Pressure24MP, temperature50℃, the rate flow of CO225L· h-1,95%ethanal (contrast to maxrix2:1) serve as entrainer. The rate of Tannin extraction was up to89.27%. The contant of Tannin was3.62%.The contant of Tannin was up to21.45%. |
