Background and objectiveIn recent years, the incidence and mortality of hepatocellular carcinoma showed anincreasing trend, The important cause of hepatocellular carcinoma treatment failure isthat it’s pathogenesis is unclear.there have been some more research about Wntsignaling pathway in hepatocellular carcinoma,But the relationship of canonical Wntsignaling pathway,non-canonical Wnt signaling pathway and GPC-3in hepatocellularcarcinoma remains unclear.In this paper, we detected the pathogenesis of Wnt5a,β-catenin and GPC-3expression in hepatocellular carcinoma and its adjacent tissues toto investigate the mechanism and to provide a new direction for targeted therapy ofhepatocellular carcinoma.Materials and methods60cases of patients with HCC specimens and the surrounding liver tissue fromtumor tissues selected from department of hepatobiliary surgery of the second artillerygeneral hospital PLA.All samples were4%formalin-fixed, paraffin-embedded,4umthick serial sections, used to prepare confirmed by histopathology andimmunohistochemistry.we analyzed the relationship of themselves and their expressionand clinicopathological parameters in hepatocellular carcinoma.Results1. The positive expression rate of Wnt5a in HCC tissues was35%(21/60), and71.7%(43/60) in adjacent tissues of Hepatocellular, compared the differences hasstatistically significance (P <0.01).2. Beta-catenin in adjacent tissues of hepatocellular positive rate was21.7%(13/60),in HCC tissues was58.3%(35/60) positive rate rise, compared the difference hasstatistical significance (P <0.05). 3.GPC-3in adjacent tissues of hepatocellular positive rate was16.7%(14/60),,inHCC tissues was88.3%(53/60)positive rate rise, compared the difference has statisticalsignificance (P <0.05).4.In hepatocellular cancer tissues, positive expression rate of Wnt5a and histologicclassification its positive expression decrease with age, sex, tumor size, histologicalgrade, presence of cirrhosis of the liver, and no significant correlation (P>0.05),andSerumAFP level (P <0.05), and TNM staging (P <0.01),statistically significant.5.In hepatocellular cancer tissues, Beta-catenin higher expression and histologicclassification statistical analysis of the abnormal expression and age, sex, tumor size,the serum AFP value, tumor TNM staging, and no significant correlation (P>0,05),histological grade, presence of cirrhosis of the liver (P <0.05), statistically significant.6.In hepatocellular cancer tissues, GPC-3higher expression and age, gender, TNMstaging, no significant correlation (P>0.05),and the relativity of Serum AFP level,tumor size, histological grade, presence of cirrhosis of the liver (P <0.05).7.In hepatocellular cancer tissues, Wnt5a protein expression and beta-catenin wasfound to be inversely associated with the abnormal expression of protein,Wnt5a proteinexpression and GPC-3protein abnormal expression showed a negative correlation,Beta-catenin protein expression and GPC-3the abnormal expression of protein werepositively correlated.Conclusion1.Wnt5a play a inhibiting role in the occurrence and development of hepatocellularcancer.2.β-catenin could be an indicator for early detection of hepatocellular cancer, butwhether it can become a target for the treatment of hepatocellular cancer, needs furtherstudy.3.GPC-3is expected to become a new target for the treatment of hepatocellularcancer because it involved in hepatocellular carcinoma occurs and the entire process ofdevelopment.4.GPC-3may be to promote tumorigenesis by binding with Wnt5a in thedevelopment of hepatocellular carcinoma. 5.Non-canonical Wnt signaling pathway represented by Wnt5a may inhibittumorigenesis by inhibiting the canonical Wnt signaling pathway.
